电针五脏俞治疗PCPA诱导大鼠失眠的疗效观察

Therapeutic Effect of Electroacupuncture at Wuzangshu on PCPA-induced Insomnia in Rats

目的:比较电针五脏俞刺激与常规西药治疗PCPA致失眠大鼠的疗效并探讨其作用机制。方法:建立大鼠PCPA失眠模型,分别采用电针五脏俞及西药地西泮给予治疗,监测大鼠治疗期间行为学与体质量变化,治疗结束后试剂盒检测血清SOD活性及MDA含量,TUNEL检测神经元凋亡,RT-PCR及Western blot检测海马NeuN、BDNF的mRNA及蛋白表达。结果:相较PCPA模型组,电针五脏俞刺激与西药地西泮处理能够显著促进失眠大鼠海马区BDNF的合成,增加脑内能正常表达NeuN的神经元数目,减少神经元细胞凋亡,提高血清SOD活性,下调MDA含量以缓解PCPA造成的机体氧化与抗氧化系统紊乱,使大鼠外在精神面貌愈发转好、体质量增速愈发恢复,从而达到治疗失眠的目的。结论:电针五脏俞刺激能对失眠大鼠大脑发挥明显的修复损伤和保护作用,其促进体内生理功能、改善中枢神经活动的效果好于常规西药地西泮或与其相当,具有较强的临床实用性。

Objective:To compare the efficacy of electro-acupuncture Wuzangshu stimulation and conventional western medicine in treating PCPA-induced insomnia in rats,and to explore its mechanism.Methods:PCPA insomnia model was established in rats.The rats were treated with electro-acupuncture at Wuzangshu and diazepam.The changes of behavior and body weight during the treatment were monitored.Serum SOD activity and MDA content were measured by reagent kit after the treatment.Neuron apoptosis was detected by TUNEL.Detection of mRNA and protein expression of Neu N and BDNF in hippocampus was measured by RT-PCR and Western blot.Results:Compared with PCPA model group,electroacupuncture Wuzangshu stimulation and diazepam treatment could significantly promote the synthesis of BDNF in hippocampus of insomnia rats,increase the number of neurons expressing Neu N normally in brain,reduce neuron apoptosis,and increase the activity of SOD in serum and decrease the content of MDA in order to alleviate the disorder of oxidation and antioxidant system caused by PCPA.Therefore,the external mental status of rats is becoming better and the weight gain is more and more recovered,so as to achieve the purpose of treating insomnia.Conclusion:Electro-acupuncture Wuzangshu stimulation can obviously repair and protect the brain of insomniac rats.Its effect of promoting physiological function and improving central nervous activity is better than that of diazepam or equivalent to it,which has strong clinical practicability.
