枸杞多糖对大鼠肾缺血-再灌注后血清中TNF-α和IL-1β含量的影响 被引量:2

Effect of lycium barbarum polysaccharides on expression of TNF-α and IL-1β in serum after ischemia-reperfusion injury of rat kidney

摘要 目的探讨缺血-再灌注对肾组织的损伤以及枸杞多糖(LBP)的治疗作用。方法实验大鼠被分成3组:LBP治疗组、假手术组和模型组,每组10只。LBP治疗组:大鼠在进行手术的前10天,每只大鼠按体重口服LBP,200 mg/kg,每天1次,以相同方式假手术组和模型组口服同等量的生理盐水。手术后48 h取大鼠肾组织,苏木素-伊红染色,显微镜下观察肾组织的结构;测各组大鼠尿量、血肌酐、尿素氮的含量的变化;酶联免疫检测大鼠血清中肿瘤坏死因子-α(TNF-α)及白介素-1β(IL-1β)的含量。结果在模型组的大鼠肾脏切片中观察到损伤及坏死,尿量[(4.35±0.51)ml]比假手术组明显减少,血浆中肌酐[(146.79±11.94)μmol/L]、尿素氮[(43.11±2.72)mmol/L]、TNF-α[(1 786.48±151.21)ng/L]及IL-1β[(1476.33±125.62)ng/L]水平比假手术组明显升高(P<0.05);提前口服LBP后大鼠肾脏组织损伤及坏死减少,尿量(7.12±0.68 ml)比模型组明显增加,血浆中肌酐[(67.83±6.98)μmol/L]、尿素氮[(14.21±0.69)mmol/L]、TNF-α[(894.32±98.76)ng/L]及IL-1β[(623.58±72.45)ng/L]水平比模型组明显减少(P<0.05)。结论缺血-再灌注会引起大鼠肾组织损伤,LBP治疗后会降低炎性因子TNF-α及IL-1β的含量来改善其损伤作用。 [Objective]To explore the ischemia-reperfusion damage in rat renal tissue and the therapeutic effect of lycium barbarum polysaccharide(LBP).[Methods]All SD rats were randomly assigned into 3 groups:LBP treament group,sham operation group and model group,10 in each group.LBP group:10 days before the operation,200 mg/kg,1 time a day were taken orally by each rat according to body weight.The same amount of normal saline was taken orally by the sham operation group and the model group in the same manner.After 48 h of ischemia reperfusion,hematoxylin-eosin staining was performed to observe the structure of renal tissue under microscope.The content of serum creatine(SCr),blood urea nitrogen(BUN)and the urine volume were detected,enzyme-linked immune was used to detect the content of tumor necrosis factor-alpha(TNF-α),interleukin-1 bate(IL-1β)in rat se rum.[Results]In the model group,injury and necrosis were observed in kidney sections of rats,the volume of urine[(4.35±0.51)ml]reduced,compared with sham operation group,the content of Scr[(146.79±11.94)μmol/L)],BUN[(43.11±2.72)m mol/L],TNF-ɑ[(1 786.48±151.21)ng/L]and IL-1β[(1 476.33±125.62)ng/L]were significantly increased(P<0.05);In the LBP treatment group,the damage and necrosis in kidney tissue decreased,the volume of urine[(7.12±0.68)ml]increased,the content of Scr[(67.83±6.98)μmol/L],BUN[(14.21±0.69)mmol/L],TNF-α[(894.32±98.76)ng/L]and IL-1β[(623.58±72.45)ng/L]were significantly decreased(P<0.05),compared with model group.[Conclusion]Ischemia-reperfusion can cause renal tissue injury in rats,LBP after treatment can reduce inflammatory factor of TNF-ɑand IL-1βcontent to improve its damage.
