摘要
目的探讨黄芪颗粒对慢性阻塞性肺疾病(COPD)模型大鼠炎性因子及血管内皮功能的影响。方法清洁级雄性SD大鼠80只,采用随机数字表法将SD大鼠分为正常对照组、模型组、黄芪低剂量组、中剂量组和高剂量组,20只/组。除正常对照组外,其他各组使用单纯香烟烟熏法构建COPD大鼠模型,于停止烟熏当天,正常对照组和模型组均灌胃给药0.9%氯化钠溶液;黄芪低剂量组、中剂量组和高剂量组分别灌胃给药黄芪颗粒0.4 g/kg、0.8 g/kg、1.6 g/kg。实验结束时,检测各组大鼠肺功能;肺组织匀浆中白介素-6(IL-6)、白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)含量;血清一氧化氮(NO)和内皮素(ET);Western-blot检测肺组织Cx43、Cav-1、e NOS、Rho A和ROCK1蛋白表达水平。结果试验黄芪低剂量组、中剂量组和高剂量组FEV0.3/FVC、血清NO水平均显著高于模型组(P <0.05),且随着黄芪颗粒用药剂量的增加,FEV0.3/FVC、血清NO水平显著上升(P <0.05)。试验黄芪低剂量组、中剂量组和高剂量组肺组织中IL-6、IL-8和TNF-α表达水平均显著低于模型组(P <0.05),且随着黄芪颗粒用药剂量的增加而显著。模型组、黄芪低剂量组、中剂量组和高剂量组Cx43、Cav-1、Rho A和ROCK1蛋白表达水平显著高于对照组(P <0.05),但显著低于模型组(P <0.05)。结论黄芪颗粒可调节COPD大鼠ET与NO平衡,改善血管内皮功能障碍,减轻气道炎症反应,这个过程可能是通过Rho/Rho激酶信号转导通路实现的。
Objective To explore the effect of Huangqi granule on inflammatory factors andvascular endothelial function in chronic obstructive pulmonary disease(COPD) model rats. Methods Eighty cleanmale SD rats were randomly divided into normal control group, model group, Huangqilow dose group, middle dose group and high dose group with 20 rats in each group. In addition to the normal control group, the other groups were used to establish the COPD rat model with the simple cigarette smoking method. On the day of cessation of smoking, 0.9% sodium chloride solution was administered orally to the normal control group and the model group. Huangqi low dose group,middle dose group and high dose group were given by Huangqi granule 0.4 g/kg, 0.8 g/kg, 1.6 g/kg. At the end of the experiment, the lung function of each group was detected. The levels of interleukin-6(IL-6), interleukin-8(IL-8) and tumor necrosis factor-α(TNF-α) were detected in the homogenate of lung tissue. Serum nitric oxide(NO)and endothelin(ET) weredetected. Western-blot was used to detect the expression of Cx43, Cav-1, eNOS, RhoA and ROCK1 protein in lung tissue. Results The levels of FEV0.3/FVC and serum NO in the Huangqilow, middle and high dose groups were significantly higher than those in the model group(P < 0.05). With the increase of the dosage of Huangqi granule, the serum NO level and FEV0.3/FVC increased significantly(P < 0.05). The expression of IL-6,IL-8 and TNF-α in lung tissue of Huangqi low dose group, middle dose group and high dose group were significantly lower than that of model group(P < 0.05), and with the increase of the dosage of Huangqi granule, it is significant.The expression levels of Cx43, Cav-1, RhoA and ROCK1 in model group, Huangqi low dose group, middle dose group and high dose group were significantly higher than those in control group(P < 0.05), but significantly lower than those in model group(P < 0.05). Conclusion Huangqi granule can regulate the balance between ET and NO,improve vascular endothelial dysfunction and alleviate airway inflammation in COPD rats. This process may be realized by Rho/Rho kinase signal transduction pathway.
