TNF-α对大鼠肾髓袢升支粗段中PKA及其磷酸化水平蛋白表达的影响

Influence of Tumor Necrosis Factor- α on the Expression of Protein Kinase A and Phosphorylation Level of Renal Thick Ascending Limb of Rat

目的:探讨肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)对大鼠肾髓袢升支粗段中PKA(蛋白激酶A)及其磷酸化水平蛋白表达的影响。方法:在髓袢升支粗段组织中加入10n M的TNF-α作用5min,采用免疫印迹技术观察TNF-α对PKA及其磷酸化水平蛋白表达的影响。结果:TNF-α未能明显改变大鼠肾髓袢升支粗段中PKA及其磷酸化水平的蛋白表达(P﹥0.05)。结论:TNF-α不能改变PKA的蛋白合成及其磷酸化水平的表达。

Objective: To study the effect of TNF- α on the protein expression of PKA and phosphorylation level in the thick ascending limb. Methods: The TNF- α was placed into the tissue of TAL for five minutes to observe the protein expression of PKA and phosphorylation level by using Western blot technique. Results: TNF- α did not change the protein expression of PKA and phosphorylation level in the thick ascending limb( P ﹥ 0. 05). Conclusion: The TNF- α can not change the protein expression of PKA and phosphorylation level.
