摘要
前列腺素I2(PGI2)具有很强的扩血管及抗血小板聚集作用,广泛地存在于血管壁及其它组织中,不稳定,在血液中半衰期仅为2~3min,难以精确定量。6-酮-前列腺素F1α(6-Keto-PGF1α)是PGI2的较稳定的非酶裂解产物,Salmon首先建立了6-Keto-PGF1α放射免疫分析法进行PGI2的定量。国外已有商用测定盒供应。我们建立了此方法,并开始应用于科研工作。
Prostacyclin(PGI2)is an important substance for vascular dilatation and platelet aggregation,but it is very easily destroyed, so that it is difficult to assay this compound in biological system. PGI2, within only 2 minutes,is metabolized to 6-keto-PGF1a.Radioimmunoassay for 6-keto-PGF1a was established in our laboratories. The cross reactivity between the antiserum and several PGs was very low,the affinity constant of the antisera being 1.48×109 L/mol,and the analytical working range being 12.5 pg to 800 pg per tube.The samples were extracted with petroleum,ether and acetic acid.resu-spended in PBS and quantitated by radioimmunoassay.The recovery of the extract was 98%,the plasma value of 6-keto-PGF1a being 138±77.9 pg/ml in normal subjects. This method has been used in measuring 6-keto-PGF1a concentration of kidney and plasma in normal and SHR/N rats.
