摘要 肿瘤耐药是限制目前肿瘤治疗众多化疗方案疗效的基本问题。耐药可以通过各种机制产生,如:增加药物失活、增加肿瘤细胞的药物外排、增强化疗导致的损伤修复、激活促生存信号通路和失活细胞死亡通路。理解肿瘤细胞耐药的产生机制有助于在临床中更好地应用化疗药物以及促进中西医对肿瘤耐药性的研究。本文回顾了肿瘤细胞耐药的一些主要机制,并进一步讨论了目前正在使用的有助于克服耐药的一些新的分子靶向治疗手段。 Drug resistance is a fundamental problem that limits the effectiveness of chemotherapies currently used to treat cancer. Drug resistance can be produced through a variety of mechanisms, such as increased drug inactivation and drug efflux from cancer cells, enhanced repair of chemotherapy-induced damage, activation of pro-survival pathways and inactivation of cell death pathways. Understanding the mechanisms of drug resistance of tumor cells contributes tothebetter application of chemotherapy drugs in clinical practice and better study of tumor drug resistance by bothTraditional ChineseMedicine and Western medicine. This paper reviews some of themain mechanisms of drug resistance of tumor cells and further discusses new molecularly- targeted therapies currentlyapplied to overcome these resistance mechanisms.
