摘要
目的:总结及探讨RH Rh(D)阴性血冰冻红细胞的制备和检测方法。方法:取1U/袋的Rh Rh(D)阴性全血红细胞,采用电子摇摆称和输血器自动控制器法控制甘油加入速度。经水浴溶融化后,对冰冻红细胞进行两次洗涤,采用GPO-PAP法测定残余甘油、邻-甲联苯胺法检测上清游离血红蛋白,并计算其他指标。结果:甘油残余量6.81±2.76 g/L,上清液游离Hb0.78±0.02(g/L),体外溶血率9.96±1.24%,红细胞回收率87.68±2.89%,均要优于国家标准。结论:采用上述方法制备、融化、洗涤、检测冰冻红细胞获得了比较满意的红细胞回收率,各项质量指标也优于国家标准。
To summarize and discuss the preparation and detection methods of RHRh(D) negative blood frozen red blood cells. . Methods: Full red blood cells of 1 u/bag of negative Rh Rh(D) was taken. Electronic swing and donor set automatic controller controlled the glycerin to join. Frozen red blood cells were washed twice after bath solution, the GPO- PAP method measured residual glycerin, and O- methyl benzidine method detected the free hemoglobin and other indicators were calculated. Results: Glycerol residual amount was 6.81 + 2.76 g/L, supernatant free Hb0.78 + /- 0.02 (g/L), in vitro hemolysis rate 9.96 + /- 1.24%, erythrocyte recycling rate 87.68 + /- 2.89%, betterthan the national standards. Conclusion: The method above in application of melting, washing, detecting frozen erythrocyte obtains satisfactory erythrocyte recovery rate and the quality indicators are better than the national standards.
