摘要
目的 :研究神经母细胞瘤中多药耐药 (MDR1)基因和谷胱甘肽转移酶 π(GST π)蛋白表达及其临床病理意义。方法 :采用S P免疫组化方法检测 72例神经母细胞瘤组织中MDR1和GST π蛋白表达 ,分析其表达与神经母细胞瘤临床病理特征的关系 ,探讨二者之间表达的关系。结果 :MDR1和GST π蛋白在神经母细胞瘤中阳性表达率分别为 62 .5% (45/72 )和 58.3 3 % (42 /72 )。MDR1蛋白表达与神经母细胞瘤的组织学分型和转移密切相关(P <0 .0 5) ;GST π蛋白表达与神经母细胞瘤的肿块大小和组织学分型密切相关 (P <0 .0 5) ;在神经母细胞瘤中MDR1和GST π蛋白表达呈正相关 (P <0 .0 5)。结论 :MDR 1和GST π蛋白可以作为反映神经母细胞瘤分化与耐药关系的分子基础 。
Objective: To investigate the expressionsofmultidrug resistance 1(MDR1)and glutathione S-transferase-π(GST -π) protein and to explore their clinicopathological significance in neuroblas toma.Methods: Seventy-two cases of neuroblastoma were studiedwith S-P immunohistochemistry using antibodies against MDR1 and GST-π. Relat ionship between the expressions and clinicopathological features of neurobla stoma, and relationship between expression of MDR1 and that of GST-π were inve stigated. Results: The positive expression rates of MDR1 and G ST-π were 62.5%(45/72)and 58.33% (42/72) in neuroblastoma, respectively.Expression of MDR1 was correlated with histological classification and metasta sis of neuroblastoma(P<0.05).Expression of GST-π was correlated with tu mor size and histological classification of neuroblastoma(P<0.05).Express ion of MDR1 was positively paralleled with that of GST-π in neuroblastoma(P < 0.05 ).Conclusion: Expressions of MDR1 and GST-π playan important role in molecular mechanism of multidrug resistance of neuroblasto ma and are associated with biological behaviors of neuroblastoma. Observation onexpressions ofMDR1 and GST-π is helpful in chemotherapy of neuroblastoma.
