摘要
目的:建立10个不同批次枫香脂鲜料HPLC指纹图谱,并测定肉桂酸的含量。方法:应用HPLC建立其指纹图谱,按"中药指纹图谱相似度评价系统(2004A版)"建立对照图谱并进行相似度评价,利用SPSS 21.0软件进行聚类分析;运用UPLC-Q-TOF/MS对其化学成分定性分析,建立肉桂酸含量测定方法。结果:建立了枫香脂鲜料HPLC指纹图谱,确立了6个共有峰,指认了4个化合物,聚类分析将其分为2类。10批样品除S4、S5、S9相似度偏低外,其余均大于0.950。经方法学验证,肉桂酸在1.632~16.320μg·ml^(-1)(r=0.999 2)内线性关系良好;平均回收率为97.83%(n=9)。结论:该文所建立的HPLC指纹图谱及肉桂酸含量测定方法,可用于枫香脂鲜料药材质量控制。
Objective: To establish the HPLC fingerprints of 10 different batches of Resina Liquidambaris fresh material,and determine the content of cinnamic acid. Methods: The software Similarity Evaluation System for Chromatographic Fingerprint of TCMs (Version 2004 A) was employed to generate the mean chromatogram and carry out the similarity analysis of the samples. Cluster analysis was used in date analysis by SPSS software (SPSS 21.0). Ultra-performance liquid chromatography quadrupole time-of-flighttandem mass spectrometry (UPLC-Q-TOF/MS) was used for the qualitative analysis of Resina Liquidambaris fresh material,and quantitative method of cinnamic acid was established.Results: The HPLC fingerprints of Resina Liquidambaris fresh material were established,among them,6 common peaks were found,and 4 compounds were identified and classified into two classes by the clustering analysis.The similarity of the 10 batches of samples,except for S4,S5 and S9 with low similarity,was more than 0.950. According to the methodological verification,cinnamic acid showed good linear relationship (r = 0.999 2) within the range of 1.632-16.320 μg·ml-1,and the average recovery was 97.83% (n = 9). Conclusion: The HPLC fingerprint and quantitative method of cinnamic acid established in the paper can be used for the quality control of Resina Liquidambaris fresh material.
