摘要
以HPLC为研究手段,应用正交实验、方差分析等方法对提取时间、加热回流温度、甲醇-0.4%磷酸比例进行优化,并以金花茶叶中的山奈酚为研究对象,建立不同种类的金花茶叶中山奈酚的定量方法。经过单因素和正交试验得出加热回流提取金花茶叶中山奈酚的最佳工艺为:加热温度为80℃,加热时间为100 min,甲醇磷酸比例为1:4。研究表明,该方法可快速准确对金花茶叶中山奈酚质量进行定量,且方法简单、灵敏度高、线性范围广。
HPLC was as the research method to optimize heat temperature.Heat time and Methanol-0.4%phosphoric acid ratio were optimized by orthogonal and variance analysis.Taking kaempferol in Camellia as study to objective set up mass spectral measurement of quality for the kaempferol.Through single factor and orthogonal experiments the optimal conditions of soaking kaempfero were determined as follows:heat extraction temperature at 80℃,extraction time of 100 min and the Methanol-0.4%phosphoric acid ratio with 1:4.The result showed that this method can realize the fast quantitative of quality mass for kaempferol in single sample with easy method,fast speed,high sensitivity and wide linear range and can be used to fast quantitative quality mass of kaempferol.
