摘要
目的研究北沙参茎叶醇提物及其不同极性部位萃取物的体外抗氧化活性。方法用95%乙醇回流提取制备北沙参茎叶醇提物,采用不同极性有机溶剂萃取北沙参茎叶醇提物,得到石油醚相、乙酸乙酯相、正丁醇相和水相4个不同极性部位萃取物。以维生素C(Vc)为对照,研究各萃取物清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基能力及其总还原能力。结果北沙参茎叶醇提物及其不同极性部位萃取物对DPPH自由基、ABTS自由基有不同程度的清除作用;在0~0.1 mg·mL-1浓度范围内,北沙参茎叶醇提物及乙酸乙酯部位、正丁醇部位对DPPH、ABTS自由基具有较强的清除作用,且均强于Vc。乙酸乙酯部位还原力最强,正丁醇部位、醇提物次之。结论北沙参茎叶醇提物及其不同极性部位萃取物具有一定的抗氧化及清除自由基的作用,可为北沙参茎叶开发利用提供新途径。
Objective To study in vitro antioxidant activity of ethanol extract of stems and leaves of Glehnia littoralis and the different polarity fractions.Methods The 95%ethanol extract was prepared from the stems and leaves of Glehnia littoralis,and different polarity fractions were prepared by extracting orderly using petroleum ether,ethyl acetate,n-butanol and water from the ethanol extract of Glehnia littoralis.The abilities of scavenging DPPH free radicals,ABTS free radicals and total reducibility were studied.Results The ethanol extract from the stems and leaves of Glehnia littoralis and their different polarity fractions can scavenge DPPH free radicals and ABTS free radicals with different degrees.Within the range of 0-0.1 mg·mL-1,the ethyl acetate fraction and n-butanol fraction of the ethanol extract of the stems and leaves of Glehnia littoralis have scavenging effects on DPPH and ABTS free radicals that stronger than Vc.Ethyl acetate had the strongest reducing power,followed loy n-butanol and ethanol extract.Conclusion The ethanol extract of stems and leaves of Glehnia littoralis and its different polarity fractions showed certain antioxidant and scavenging effects on free radicals.This provided new direction for the development and utilization of stems and leaves of Glehnia littoralis.
