和厚朴酚改善2型糖尿病KK/Upj-AY小鼠糖脂代谢的药效学研究 被引量:7

Pharmacodynamic Study of Honokiol on Improving Glucose and Lipid Metabolism in KK/Upj-AY Mice with Type 2 Diabetes Mellitus

摘要 目的研究和厚朴酚对2型糖尿病KK/Upj-AY小鼠的降血糖作用、血脂各项指标的变化以及对胰岛组织的影响。方法 KK/Upj-AY小鼠给予高脂饲料复制2型糖尿病模型,通过灌胃给药35 d,观察各组小鼠的相应检测指标的差异和胰腺组织病理切片。结果与正常组比较,模型组小鼠的血糖值、药时曲线下面积(AUC)、胰岛素表达水平、体质量均显著升高(P<0.01);模型组小鼠血清中胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)及高密度脂蛋白(HDL)含量变化均无统计学意义;模型组小鼠胰腺平均胰岛数显著增多(P<0.01)。与模型组比较,从第1周开始,和厚朴酚中、高剂量组及吡格列酮组小鼠的血糖值均显著降低(P<0.05,P<0.01);和厚朴酚中、高剂量组及吡格列酮组小鼠的AUC值均显著降低(P<0.05,P<0.01);和厚朴酚中、高剂量组及吡格列酮组小鼠的胰岛素表达水平显著降低(P<0.01);和厚朴酚低剂量组小鼠血清中TC及TG含量显著降低(P<0.01);和厚朴酚中剂量组小鼠血清中TG、TC及LDL含量均显著降低(P<0.05,P<0.01);和厚朴酚高剂量组小鼠血清中TC含量显著降低,HDL含量显著升高(P<0.05,P<0.01);吡格列酮组小鼠血清TG及LDL含量均显著降低(P<0.01);和厚朴酚中剂量组小鼠的体质量从第2周开始显著降低(P<0.01),和厚朴酚高剂量组小鼠的体质量从第1周开始显著降低(P<0.05),吡格列酮组小鼠的体质量从第2周开始显著降低(P<0.05);和厚朴酚高剂量组的平均胰岛面积显著增大(P<0.05)。结论和厚朴酚具有降低2型糖尿病KK/Upj-AY小鼠血糖值的作用,还具有改善血脂紊乱导致的胰岛素抵抗作用,同时没有诱导小鼠体质量增加。 Objective To study the effects of honokiol on hypoglycemic,blood lipids and islet tissue in type 2 diabetic KK/UPj-AY mice.Methods The type 2 diabetes models were produced by giving KK/UPj-AY mice high-fat feed.The difference of the detection indexes and pathological sections of pancreatic tissue of mice in each group were observed throughout the 35 days’gastric administration.Results Compared with the normal group,the blood sugar value,drug time curve area(AUC)value,insulin expression level and body mass of the mice in the model group all increased significantly(P<0.01).The serum levels of TC,TG,LDL and HDL were not significant different.The mean number of pancreas islets increased significantly in the model group.Compared with the model group,from the first week,the blood sugar values of mice in the medium,high dose groups and the pioglitazone group were significantly reduced(P<0.05,P<0.01);the AUC were significantly decreased in the medium and high dose groups and the pioglitazone group(P<0.05,P<0.01).The expression of insulin decreased significantly in the middle,high dose groups and the pioglitazone group(P<0.01).The levels of TC and TG in the serum of mice with low dose of honokiol decreased significantly(P<0.01).The levels of TG,TC and LDL in serum were significantly decreased in the middle dose group(P<0.05,P<0.01).The level of TC decreased significantly and that of HDL increased significantly in the serum of mice treated with high dose of honokiol decreased(P<0.01,P<0.05).The serum TG and LDL in the pioglitazone group were significantly decreased(P<0.01).The body mass of mice in the medium dose group of honokiol decreased significantly from the second week(P<0.01),and the body mass of mice in the high dose group of honokiol decreased significantly from the first week(P<0.05).The body mass of the mice in the pioglitazone group decreased significantly from week 2(P<0.05,P<0.01).The average islet area increased significantly in the high dose group mice(P<0.05).Conclusion Honokiol could decrease blood sugar of KK/Upj-AY mice with type 2 diabetes mellitus,but did not induce mass gain.It can also improve insulin resistance produce by blood lipid disorders.
