摘要
目的:研究苍术与苍术素在大鼠体内药代动力学的差异。方法:采用反相高效液相色谱法测定苍术水提液中苍术素及苍术素单体大鼠的血药浓度,采用DAS计算药动学参数。结果:苍术水提液中的苍术素在大鼠体内的达峰时间Tmax为62.32±13.26 min、最大浓度Cmax为28.26±6.37 ng/ml、半衰期T1/2166.36±21.22min、AUC0-∞为2748.54±226.3ng/ml.min;苍术素在大鼠体内的达峰时间Tmax为37.28±9.23min、最大浓度Cmax为17.83±4.65 ng/ml、半衰期T1/2为96.48±18.34 min、AUC0-∞为2065.42±206.51ng/ml.min。苍术水提液组和苍术素单体组的主要药动学参数的差异具有显著性统计学意义。结论:苍术水提液中苍术素及苍术素单体在大鼠体内药动学特征有显著影响,提示苍术水提液中其他成分会影响苍术素在大鼠体内的药动学行为。
Objective: To Comparative pharmacokinetics of atractylodin between pure atractylodin and Atractylodes lancea(Thunb.) DC.in rats.Methods: The concentrations of atractylodin in rat plasma were determined by HPLC method.The pharmacokinetic parameters were calculated using DAS and t test was used for statistical analysis.Result: Results After oral administration of pure atractylodin and Atractylodes lancea(Thunb.) DC.Atractylodes lancea(Thunb.) DC: the Tmax was 62.32±13.26 min,Cmax was 28.26±6.37 ng/ml,T1 /2was 166.36 ±21.22 min,AUC was 2748.54±226.3ng/ml.min.Pure atractylodin: the Tmax was37.28±9.23 min,Cmax was 17.83±4.65 ng/ml,T1 /2was 96.48±18.34 min,AUC was 2065.42±206.51 ng/ml.min.The mean Cmax,AUC0-∞,tmax different than those administrated pure atractylodin.Conclusion: The chemical-chemical interactions may be accounting for the different pharmacokinetic behaviors of atractylodin after oral administration of pure atractylodin and Atractylodes lancea(Thunb.) DC.in rats.
