鸡骨香醇提物的抗炎作用 被引量:7

Anti-inflammatory effects and possible mechanism of action of Croton crassifolius ethanol extract

摘要 目的:研究鸡骨香醇提物的抗炎作用。方法:采用二甲苯至致耳肿胀、角叉菜胶致大鼠足肿胀、琼脂致大鼠肉芽肿实验观察鸡骨香的抗炎作用;测定角叉菜胶致胸膜炎大鼠胸腔渗出液白细胞数量、前列腺素E2、肿瘤坏死因子-α和白细胞介素-6的含量,探讨鸡骨香抗炎机制。结果:鸡骨香醇提物180 mg/kg、90 mg/kg能抑制二甲苯致小鼠耳肿胀;醇提物三个剂量能抑制角叉菜胶致炎6 h大鼠足肿胀和琼脂致大鼠肉芽肿增生;醇提物180 mg/kg能抑制胸膜炎大鼠胸腔渗出液白细胞募集、显著降低渗出液前列腺素E2、肿瘤坏死因子-α和白细胞介素-6水平;醇提物90 mg/kg能降低胸腔渗出液前列腺素E2、肿瘤坏死因子-α水平。结论:鸡骨香具有一定的抗炎作用,机制可能与影响炎症介质表达有关。 Objective: To observe the anti-inflammatory effects of Croton crassifolius ethanol extract,and explore its mechanism.Methods: Antiinflammatory effects were determined by the models of xylene-induced ear swelling in mice,carrageen-induced edema of the hind paw and agar-induced granuloma in rats.The anti-inflammatory mechanism was investigated by detecting the number of leukocytes and the contents of prostaglandins E2,tumor necrosis factor-alpha and interleukin-6 in carrageen-induced pleurisy rats pleural exudates.Results: C.crassifolius ethanol extract 180 mg/kg,90 mg/kg showed the inhibitory effect on xylene-induced ear swelling in mice,ethanol extract of three doses suppressed edema of the hind paw in rats at 6 h after carrageenin injection and agar-induced granuloma in rats,ethanol extract 180 mg/kg decreased the total leukocytes in the rats exudates,and also significantly reduced the levels of prostaglandins E2,tumor necrosis factor-alpha and interleukin-6,the extract 90 mg/kg reduced the contents of prostaglandins E2 and tumor necrosis factor-alpha in the exudates.Conclusion: C.crassifolius presents significant anti-inflammatory effects,which may be the result of inhibition of inflammatory mediators expression.
