摘要
目的:研究白芍甘草(炙)药对对原发性痛经模型大鼠谷胱甘肽过氧化物酶(GSH-Px)、活性氧(ROS)、4-羟基壬烯酸(4-HNE)、8-羟基脱氧鸟苷(8-OHd G)、前列腺素E2(PGE2)、6-酮-前列腺素(6-K-PGF1α)的影响,确定白芍甘草(炙)药对最佳配伍比例。方法:采用腹腔注射己烯雌酚与缩宫素制备原发性痛经模型,观察空白对照组、模型组、白芍组、甘草组、白芍甘草1:1、2:1、3:1组扭体次数;采用酶联免疫吸附法测定血清GSH-Px、ROS、4-HNE、8-OHd G、PGE2、6-K-PGF1α水平;剖取子宫,制备10%匀浆,测定PGE2、6-K-PGF1α活性。结果:针对大鼠血清,模型组各指标含量ROS为289.1±3.6 IU/ml、GSH-Px为33.3±0.5 pmol/ml、4-HNE为342.0±1.9 pg/ml、8-OHd G为71.8±1.0 ng/L、PGE2为419.7±5.7 ng/L、6-K-PGF1α为60.7±1.1 ng/ml,白芍甘草1:1组显著降低4-HNE、8-OHd G、PGE2含量(309.8±4.6 pg/ml、64.6±0.5 ng/L、377.0±6.0 ng/L),白芍甘草2:1组显著降低ROS、4-HNE、8-OHd G、PGE2、6-K-PGF1α含量(233.5±0.6 IU/ml、287.8±2.4 pg/ml、58.6±1.5 ng/L、372.8±1.5 ng/L、49.5±0.4 ng/m L),升高GSH-Px含量(45.7±0.5 pmol/ml),白芍甘草3:1组显著升高GSH-Px含量(42.8±1.0 pmol/ml);针对子宫匀浆,模型组PGE2、6-k-PGF1α含量为(390.0±5.6 ng/L、49.0±0.9 ng/m L)白芍甘草2:1组显著降低PGE2、6-K-PGF1α含量(363.1±4.1ng/L、37.6±0.2ng/m L),白芍甘草3:1组显著降低6-K-PGF1α含量(42.9±0.8 ng/m L),以上结果均具有统计学意义。结论:白芍甘草(炙)药对升高痛经大鼠体内GSH-Px,降低ROS,4-HNE,8-OHd G、PGE2、6-K-PGF1α含量,通过抗氧化应激及镇痛作用达到治疗效果,确定白芍甘草(炙)药对最佳配伍比例为2:1。
Objective: To investigate the effects of match-pair of Paeoniae Radix alba and Glycyrrhizae Radix et Rhizoma cum melle(S-G matchpair) about GSH-Px,ROS,4-HNE,8-OHd G,PGE2,6-K-PGF1α on primary dysmenorrhea model rats and explore the best proportion and treatment mechanism of S-G match-pair.Methods: Injected diethylstilbestrol and oxytocin into rats to get the dysmenorrheal model and to study the treatment effects of S-G match-pair(1: 0,1: 1,2: 1,3: 1,0: 1) and the control group; All rats were detected serum GSH-Px,ROS,4-HNE,8-OHd G,PGE2,6-K-PGF1α by ELISA; Obtained the living uterine tissue homogenized into homogenate,which determined the activity of PGE2,6-K-PGF1α by ELISA.Results: Compared with the mode,in serum,S-G match-pair 1: 0 significantly reduced the content of 8-OHd G,PGE2(64.9±0.7 ng/L,386.0±3.7 ng/L),S-G match-pair 0: 1reduced the content of PGE2(387.1±4.0 ng/L),S-G match-pair1: 1 significantly reduced the content of 4-HNE,8-OHd G,PGE2(309.8±4.6 pg/ml,64.6±0.5 ng/L,377.0±6.0 ng/L),S-G match-pair2: 1 significantly reduced the content of ROS,4-HNE,8-OHd G,PGE2,6-K-PGF1α(233.5±0.6 IU/ml,287.8±2.4 pg/ml,58.6±1.5 ng/L,372.8±1.5 ng/L,49.5±0.4 ng/m L) and increased the content of GSH-Px(45.7±0.5 pmol/ml),S-G match-pair 3: 1 significantly increased the content of GSH-Px(42.9±0.8 ng/m L); In Uterine tissue homogenate,S-G match-pair 2: 1 significantly reduced the content of PGE2,6-k-PGF1α(363.1±4.1ng/L,37.6±0.2ng/m L),S-G match-pair 3: 1 significantly reduced the content of 6-K-PGF1α(42.9±0.8ng/m L).The above results were statistically significance.Conclusion: The S-G match-pair increase the content of GSH-Px,reduce the content of ROS,4-HNE,8-OHd G,PGE2,6-K-PGF1α on primary dysmenorrhea model rats.The S-G match-pair can resist oxidative stress and pain and determine the best compatibility proportion is 2: 1.
