甲基化尿石素A对油酸诱导的HepG2细胞脂肪变性的抑制作用

摘要 目的:研究甲基化尿石素A(methylated urolithin A; CH3OUro A)对油酸诱导的Hep G2细胞脂肪变性的影响。方法:采用油酸诱导Hep G2细胞脂肪变性,运用CCK8细胞活力检测法、油红O染色法、三酰甘油(TG)含量测定法和ALT/AST含量测定法,评价CH3OUro A对油酸诱导的Hep G2细胞脂肪变性的作用;运用实时荧光定量PCR法检测FASN、SREBP-1、PPARα和PPARγ等基因mRNA表达,探讨CH3OUro A对油酸诱导的Hep G2细胞脂肪变性的作用机制。结果:CH3OUro A在20μM、40μM能改善油酸诱导Hep G2细胞脂肪累积;显著降低脂肪变性模型中脂质含量和TG含量;同时还能显著降低脂肪变性模型中ALT、AST的含量;显著降低脂肪变性模型中FASN、SREBP-1和PPARγ的mRNA表达水平;上调该模型中PPARα的mRNA表达水平。结论:CH3OUro A对油酸诱导的Hep G2细胞脂肪变性有一定的改善作用,该作用与调控肝细胞中FASN、SREBP-1、PPARα和PPARγ基因的表达有关。 Objective: To investigate the effect of methylated urolithin A( CH3 OUro A) on oleic acid induced hepatic steatosis in Hep G2 cells.Methods: CCK-8 assay was performed to evaluate the effects of CH3 OUro A and OA on cell viability. Lipid droplets in the cells were observed through oil red staining. The levels of TG,ALT and AST were also measured with analyzed kits. Quantitative real-time PCR was employed to estimate the mRNA expressions of FASN,SREBP-1,PPARα and PPARγ. Results: CH3 OUro A decreased oleic acid-induced lipid content,TG content and the level of ALT and AST in Hep G2 cells. CH3 OUro A down-regulated the mRNA expressions of FASN,SREBP-1and PPARγ,as well as elevated the mRNA expression of PPARα,compared to those in the model group. Conclusion: CH3 OUro A could improve the oleic acid induced hepatic steatosis in Hep G2 cells,which is involved in regulating expression of FASN,SREBP-1,PPARα and PPARγ in hepatocytes.
