摘要
目的:运用基线等比增减设计模型,探讨酒蒸黄连、石菖蒲组分抗糖尿病认知障碍的配伍规律,以期指导糖尿病临床合理用药。方法:分别建立不同糖尿病认知障碍动物模型,从学习记忆能力、糖脂代谢水平、海马神经细胞形态学变化等药效方面综合评判。结果:酒蒸黄连-石菖蒲组分不同配伍比例能明显或部分改善小鼠糖脂代谢,改善胰岛素抵抗,提高小鼠学习记忆能力,改善脑组织病理形态学改变,减少Aβ的沉积,改善认知功能障碍,其中配比4(黄连总生物碱54. 7mg∶石菖蒲挥发油4. 17mg)疗效较好,并获得药效验证。结论:酒蒸黄连-石菖蒲组分具有确切的改善糖尿病认知功能障碍作用,最佳配比为酒蒸黄连总生物碱54. 7mg:石菖蒲挥发油4. 17mg(以原药材计,酒蒸黄连0. 835g∶石菖蒲2. 5g),即配比为0. 33∶1。
Objective: To study the component compatibility regularity of Steamed Coptis chinensis with wine and Grass leaf sweetflag rhizome against diabetic cognitive impairment by using the baseline equal ratio increase-decrease design model,in order to guide the reasonable use of drugs in diabetes clinic. Methods: Different animal models of diabetic cognitive impairment were established,and their pharmaceutical effects were comprehensively evaluated from the aspects of learning and memory abilities,glucose and lipid metabolic level,morphological changes of hippocampal neurons and so on. Results: The different compatibilities of Steamed Coptis chinensis with wine and Grassleaf sweetflag rhizome obviously or partly improved glucose and lipid metabolism,insulin resistance,learning and memory abilities and pathological morphological changes of brain tissues as well as reduced the deposition of Aβ,and ameliorated cognitive dysfunction in mice. The compatibility number 4 had better curative effect. Conclusion: The components of Steamed Coptis chinensis with wine and Grassleaf sweetflag rhizome have definite effect on the improvement of diabetic cognitive impairment. The best proportion is 54. 7 mg steamed Coptis chinensis alkaloid and 4. 17 mg Grassleaf sweetflag rhizome( raw materials: 0. 835 g Steamed Coptis chinensis;2. 5g Grassleaf sweetflag rhizome),that ratio is0. 334: 1.
