摘要
目的:研究痛麻杞宁方对大鼠实验性糖尿病周围神经病变(DPN)的改善作用及机制。方法:采用高糖高脂饲料喂饲加腹腔注射链脲佐菌素复制DPN大鼠模型。造模成功后,将模型大鼠随机分为模型对照组、二甲双胍200 mg/kg组、痛麻杞宁方30g生药/kg、15 g生药/kg和7. 5 g生药/kg组,另设空白对照组,每组10只。连续灌胃给药6周后,检测各组大鼠的足迹步态和坐骨神经SNCV;取坐骨神经HE染色;腹主动脉取血检测大鼠血浆FPG、GHb水平、血清FINS、TC、TG、LDL-C、HDL-C、ET-1和NO水平、RT-PCR法测定大鼠肝细胞膜InsR和坐骨神经eNOS mRNA表达水平。结果:痛麻杞宁方30 g生药/kg组能明显增加DPN模型大鼠后足PL和TS值,明显提高大鼠坐骨神经SNCV,改善坐骨神经病变,使大鼠血浆FPG水平出现下降趋势,明显降低血浆GHb及血清FINS水平,显著降低血清TC、TG、LDL-C及ET-1水平,明显提高血清HDL-C和NO水平,显著上调肝细胞膜InsR和坐骨神经组织中eNOS mRNA的表达。结论:痛麻杞宁方对大鼠实验性DPN具有改善性作用,其作用机制可能与调节血糖、血脂水平和血管舒张功能有关。
Objective: To study the influence and mechanism of Tongma Qining Fang on model rats with diabetic peripheral neuropathy( DPN).Methods: The model rats of DPN were established by high glucose and high fat diet plus intraperitoneal injection of streptozotocin. The rats were randomly divided into six groups: the control group,the model group,Metformin suspension group( 200 mg/kg Metformin),30 g/kg Tongma Qining Fang group,15 g/kg Tongma Qining Fang group,and 7. 5 g/kg Tongma Qining Fang group,with 10 rats in each group. All rats were intragastrically administered for 6 weeks except for those in the model group and the control group. After the last administration,the footprint parameters,SNCV of sciatic nerve,the levels of FPG and GHb in plasma,the levels of FINS,TC,TG,LDL-C,HDL-C,ET-1and NO in serums were detected in each group. HE staining was used to detect the pathological alterations of Sciatic nerve tissues. Then the mRNA expression of insulin receptor in liver membrane and eNOS in sciatic nerve were detected by RT-PCR. Results: Compared with the model group,PL and TS of foot and SNCV of sciatic nerve in 30 g/kg Tongma Qining Fang group were significantly improved,the pathological alterations of Sciatic nerve tissues were alleviated,the level of FPG in 30 g/kg Tongma Qining Fang group were decreased,the levels of GHb,FINS,TC,TG,LDL-C and ET-1 were significantly decreased,the levels of HDL-C and NO were significantly increased,and the mRNA expressions of InsR and eNOS were significantly increased. Conclusion: Tongma Qining Fang can significantly improve experimental DPN in rats. Its action mechanism might be related to adjusting blood glucose,blood lipid and vasodilation function.
