通痹胶囊对高脂血症大鼠血脂和血液流变学的影响 被引量:6

Effects of Tongbi Capsule on Blood Lipids and blood rheology in Hyperlipidemia Rats

摘要 目的:观察通痹胶囊对实验性高脂血症大鼠血脂水平、血液流变学和肝功能的影响,为其临床研究及合理应用提供参考。方法:雄性SD大鼠60只,随机分为正常对照组、高脂模型组、阳性对照组(辛伐他汀片10mg/kg)和通痹胶囊186、93、46.5mg/kg组。除正常对照组外,其余各组大鼠均每天上午灌胃高脂乳剂1 ml/100g,每天下午灌胃相应药物或生理盐水,30天后腹主动脉采血,检测各鼠血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、ALT和AST水平及血液流变学指标,并计算动脉粥样硬化指数(AI),显微镜下观察肝组织形态学变化。结果:模型组大鼠较正常组血清TG、TC、LDL-C、ALT、AST水平和AI显著升高,HDL-C水平显著降低;通痹胶囊186、93g/kg组大鼠与模型组比较血清TG、TC、LDL-C水平及AI显著降低;通痹胶囊186mg/kg组大鼠血清HDL-C水平显著升高;通痹胶囊高、中、低剂量组大鼠血清LDL-C和AI水平呈剂量依赖性降低。模型组大鼠较正常组全血粘度、血浆粘度和红细胞聚集指数显著升高;辛伐他汀片组和通痹胶囊186、93g/kg组全血粘度、血浆粘度和红细胞聚集指数较模型组显著降低;模型组大鼠肝细胞脂肪变性,通痹胶囊各剂量组大鼠肝细胞脂肪变性较轻。结论:通痹胶囊对实验性高脂血症大鼠具有一定的调血脂作用和改善血液流变学的作用,并可减轻高脂血症所致的肝细胞脂肪变性。 Objective: To observe the effect of Tongbi capsule on blood lipids and blood rheology in experimental hyperlipidemia rats and provide reference for its clinical research and rational application. Methods: 60 male SD rats were randomly divided into the normal control group,high-fat model group,simvastatin group( positive control group) and high,medium and low doses of Tongbi capsule groups. Except the normal group,rats in all other groups were intragastrically treated with 1 ml/100 g high fat emulsion every morning,rats in high,medium and low doses of Tongbi capsule groups were intragastrically administered with 186 mg/kg,93 mg/kg,46. 5 mg/kg of Tongbi capsule every afternoon,rats in the positive control group were given 10mg/kg simvastatin tablets,the normal control group and model group were irrigated normal saline at the same time every day. After 30 days,blood samples were collected,serum levels of total cholesterol( TC),triglyceride( TG),low-density lipoprotein cholesterol( LDL-C),high-density lipoprotein cholesterol( HDL-C) ALT and AST were determined and blood rheology was measured,the atherogenic indexes( AI) were calculated and the morphological changes of liver tissues were observed by microscope. Results: Compared with the normal control group,serum levels of TG,TC,LDL-C,ALT and AST and AI were increased in the model group while HDL-C level was decreased. Compared with the model group,TG,TC and LDL-C levels and AI were decreased in simvastatin group,high and medium doses of Tongbi Capsuledose groups. HDL-C levels in simvastatin and high dose of Tongbi Capsule groups were increased. LDL-C levels and AI in all doses of Tongbi capsule groups were decreased in a dose-dependent manner. Compared with the control group,the whole blood viscosity,plasma viscosity and erythrocyte aggregation index of rats in the model group were increased. Compared with the model group,the above parameters in simvastatin group,Tongbi capsule high and medium doses of groups were decreased. The liver cells in the model group showed fatty degeneration,while there was no obvious change in Tongbi capsule groups. Conclusions: Tongbi capsule can regulate blood lipids,improve blood rheology and reduce hepatocyte fatty degeneration in experimental hyperlipidemia rat model.
