隐丹参酮对小鼠下肢缺血的保护作用研究 被引量:4

Effect of Cryptotanshinone on lower limb ischemia in mice

摘要 目的:探讨丹参脂溶性有效成分隐丹参酮对小鼠下肢缺血的影响。方法:选取ICR雌性小鼠,随机分为空白组、模型组、假手术组、溶剂组(PBS+4%Tween-80)、前列地尔0.01 mg/kg组、隐丹参酮10 mg/kg、20 mg/kg、40 mg/kg组。采用左下肢股动脉结扎及分支剔除术构建小鼠下肢缺血模型,其中假手术组仅进行下肢股动脉分离术。通过体式显微镜记录各组小鼠下肢缺血症状并对其进行评分,采用HE染色观察隐丹参酮对下肢缺血模型小鼠下肢肌肉病理组织以及炎症反应的影响,采用免疫组织化学的方法考察隐丹参酮对下肢缺血模型小鼠血管生成及微血管密度(microvascular density,MVD)的影响。结果:20、40 mg/kg隐丹参酮组小鼠左下肢缺血症状评分较模型组显著降低,小鼠左下肢缺血肌肉组织MVD显著增加;40 mg/kg隐丹参酮组小鼠左下肢缺血肌肉组织异常病理反应有所改善;可显著增加小鼠左下肢腓肠肌中CD34、α-SMA的表达。结论:丹参中脂溶性有效成分隐丹参酮对小鼠下肢缺血具有保护和促进血管修复的作用。 Objective:To investigate the effect of cryptotanshinone,a lipid soluble active ingredient of salvia miltiorrhiza on lower limb ischemia in mice.Methods:Female ICR mice were randomly divided into 8 groups,including the control group,the model group,the sham operation group,the solvent group(PBS+4%Tween-80),alprostadil group(0.01 mg/kg),cryptotanshinone groups(10 mg/kg,20 mg/kg,40 mg/kg).lower limb ischemia mice model was established by separation and femoral artery ligation of left leg branch.In the sham operation group,femoral artery was only exposed but not ligated.After the model was established,ischemia symptom was observed and scored through stereomicroscope.The effect of cryptotanshinone on the pathological tissue and inflammatory response of lower limb muscle in ischemia model mice was observed by H&E staining.Effects on angiogenesis and microvascular density(MVD)in ischemia model mice were observed by immunohistochemistry.Results:Compared with the model group,cryptotanshinone at doses of 20 mg/kg and 40 mg/kg significantly reduced the ischemia symptom score of the left lower limb of mice.MVD of pathological changes in ischemic muscle tissue was significantly increased.The expressions of CD34 andα-SMA in ischemic muscle tissues were significantly increased in 40 mg/kg cryptotanshinone group.The abnormal pathological changes were improved.Conclusion:Cryptotanshinone can protect the lower limb ischemia and promote its repairmen in mice.
