摘要 目的:建立龟甲胶饮片中牛皮源、驴皮源及龟甲胶成分同时检测方法。方法:采用胰蛋白酶对龟甲胶样品酶解处理,利用超高效液相色谱-串联质谱联用技术(UPLC-MS/MS)对样品中龟甲胶、黄明胶、阿胶的特征肽离子对进行检测。结果:龟甲胶、黄明胶、阿胶3种对照药材在所建方法下保留时间为3~8 min,无基质干扰,检测限分别为0.5,2.0,1.0 mg/L,且重复性好,用于18批市售龟甲胶饮片的检测分析。结论:所建方法专属性强、灵敏度高,实现了龟甲胶饮片中主成分鉴别与非法添加成分检查的同时检测,提高检验效率,可有效控制龟甲胶质量。 Objective:To develop an ultra-high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) method for simultaneously detect bovine-hide gelatin, donkey-hide gelatin and tortoise shell gelatin in tortoise shell gelatin slices for decoction. Methods:The characteristic peptide ions of tortoise shell gelatin, bovine-hide gelatin and donkey-hide gelatin were detected by UPLC-MS/MS respectively after being hydrolyzed by trypsin. Results:The retention time of tortoise shell gelatin, bovine-hide gelatin and donkey-hide gelatin was 3-8 min in this method and there was no matrix effect. The detection limits were 0.5, 2.0, 1.0 mg/L, respectively. 18 batches of tortoise shell gelatin slices were analyzed by this UPLC-MS/MS method we established successfully. Conclusion:The method has high specificity and high sensitivity, which can realize simultaneous detection of main component identification and illegally added component inspection in tortoise shell gelatin slices for decoction, improve verification efficiency and effectively control the quality of tortoise shell gelatin.
