摘要
目的观察中药双参通脉颗粒对大鼠心肌细胞凋亡的干预作用,并探讨其作用机制。方法选取健康SD大鼠60只,随机分为6组:假手术组、模型组、西药组、中药低剂量组、中药中剂量组、中药高剂量组。对模型组、中药组、西药组实验动物进行结扎左冠状动脉前降支造模,复制急性心肌缺血模型。西药组及中药组分别给予盐酸地尔硫卓片、不同剂量中药双参通脉颗粒灌胃2周。测定心肌组织病理变化、原位末端转移酶标记技术(TUNEL)染色评估心肌细胞凋亡指数,WesternBlot法检测各组心肌组织中P-JAK2的表达水平。结果与假手术组比较,模型组P-JAK2蛋白表达显著下降(P <0.05);与模型组比较,中药低剂量组及中药高剂量组P-JAK2蛋白表达均明显升高(P <0.05)。结论中药双参通脉颗粒能够抑制大鼠心肌细胞凋亡,其机制可能与上调PJAK2表达有关。
Objective To observe the effect of Shuangshen Tongmai granule(STG)on cardiomyocyte apoptosis in rats,and to explore its mechanism.Methods Sixty healthy Sprague-Dawley(SD)rats were randomly divided into 6 groups:sham operation group,model group,western medicine group(diltiazem hydrochloride tablets),STG low-dose,group,medium dose group,and high dose group.Experimental rats in the model group,STG group,and western medicine group were replicated for the model of acute myocardial ischemia.Rats were given diltiazem hydrochloride tablets in western medicine group,STG in STG group for 2 weeks.Pathological changes of myocardial tissue were determined,the apoptosis index of myocardial cells was evaluated by TUNEL staining.The expression level of P-JAK2 in myocardial tissue was detected by western blot.Results Compared with the sham operation group,P-JAK2 protein expression was significantly decreased in the model group(P<0.05).Compared with the model group,P-JAK2 expressionS were increased in all treatment groups,and P-JAK2 protein expression were significantly increased in STG low-dose group and high-dose group(P<0.05).Conclusion STG can inhibit apoptosis of myocardial cells in rats,and its mechanism may be related to up-regulation of P-JAK2 expression.
