摘要
目的探讨乳积方对乳腺癌细胞株MDA-MB-231及MCF-7体外增殖的影响。方法体外常规培养人乳腺癌细胞MDA-MB-231及MCF-7,分别加入不同浓度梯度(0.2 g/ml,0.1 g/ml,0.05 g/ml,0.025 g/ml,0.0125 g/ml,0.00 625 g/ml)的乳积方药液,处理72h。采用四氮唑蓝(MTT)比色法检测细胞增殖。倒置显微镜下观察细胞形态的变化且拍照。设不加乳积方药液常规培养为空白对照组、人乳腺正常细胞MCF-10A为阴性对照组。结果倒置显微镜下观察不同药物浓度处理后的乳腺癌细胞和正常细胞均受到了增殖抑制。将三组细胞(人乳腺正常细胞MCF-10A及人乳腺癌细胞株MDA-MB-231、MCF-7)的药物浓度与细胞存活率数据做散点图并用趋势线拟合分别得到方程y=0.015 01x-0.814 33(R2=0.873 03),y=-0.308 62 ln4.449 09x(R2=0.897 13),y=-0.351 51 ln 4.357 06x(R2=0.944 23),其中x代表药物浓度,y代表细胞存活率。计算细胞IC50值时,三种模型对应的药物浓度依次是0.013 g/ml,0.044 g/ml,0.055 g/ml。结论 MDA-MB-231细胞的IC50值低于MCF-7细胞。初步说明乳积方制剂对MDA-MB-231细胞具有更显著的增殖抑制作用。
Objective To investigate the effect of Ruji recipe on the proliferation of breast cancer cell line MDAMB-231 and MCF-7 in vitro.Methods Human breast cancer cell line MDA-MB-231 and MCF-7 were cultured in vitro for 72 h with various concentration gradients of Ruji Recipe solution of 0.2,0.1,0.05,0.025,0.0125,0.00625 g/mL.And then the proliferation of MDA-MB-231 and MCF-7 cells was detected by tetrazolium blue(MTT)colorimetry.The morphological changes of the cells were observed under inverted microscope.The breast cancer cells culturing with routine media without Ruji Recipe solution added served as blank control,and normal human breast cell MCF-10 A was used for the negative control group.Results The proliferation of breast cancer cells and normal cells treated with different drug concentrations was inhibited under inverted microscope.The data of drug concentration correlating with the cell survival rate of the human normal breast cell MCF-10 A and human breast cancer cell line MDA-MB-231 and MCF-7 were described by scatter plots,and then we obtained the fitting equation of y=0.015 01 x-0.814 33(R2=0.873 03),y=-0.308 62 ln 4.449 09 x(R2=0.897 13),and y=-0.351 51 ln 4.357 06 x(R2=0.944 23)according to the trend line,in which x refers to the drug concentration and y refers to the cell survival rate.The IC50 value for the cell model MCF-10 A,MDA-MB-231,and MCF-7 was 0.013 g/ml,0.044 g/ml and 0.055 g/mL respectively.Conclusion The IC50 of MDA-MB-231 cell is lower than that of MCF-7 cells,which indicates that Ruji Recipe has more obvious inhibitory effect on the proliferation of MDA-MB-231 cells.
