白藜芦醇白蛋白纳米粒对人胆囊癌GBC-SD细胞凋亡的影响 被引量:2

Effect of Resveratrol Bovine Serum Albumin Nanoparticles on Apoptosis of Human Gallbladder Carcinoma GBC-SD Cells

摘要 目的探讨白藜芦醇白蛋白纳米粒对体外培养的人胆囊癌GBC-SD细胞株凋亡的影响,为白藜芦醇纳米制剂的临床应用及胆囊癌的临床治疗提供新的思路。方法将实验对象分为空白对照组、阳性药物5-Fu对照组、白藜芦醇组、白藜芦醇白蛋白纳米颗粒组、空白纳米粒组,并利用CCK-8及RT-PCR评估其体外抗肿瘤活性,检测不同浓度及药物组对凋亡相关基因Caspase-3、Caspase-8、Caspase-9 mRNA表达水平的影响。结果空白纳米粒组对GBC-SD细胞的生长无抑制作用,而三种药物组在10~80μg/ml浓度区间对GBC-SD细胞的生长有抑制作用,且呈一定的浓度依赖性。与RES相比,在20~80μg/ml浓度时5-Fu与RES-BSANP对胆囊癌GBC-SD的增殖抑制率明显增强。5-Fu、RES及其白蛋白纳米制剂均能通过上调胆囊癌GBC-SD细胞中Caspase-3、Caspase-8、 Caspase-9 m RNA的相对表达量,促进胆囊癌细胞凋亡。结论白藜芦醇白蛋白纳米粒对胆囊癌GBC-SD细胞株增殖有明显的抑制作用,而且抑制作用相较于单独的白藜芦醇要更强,这种作用机制可能与上调凋亡基因Caspase-3、Caspase-8、Caspase-9有关。 Objective To investigate the effect of resveratrol(RES)bovine serum albumin nanoparticles(BSANP)on the apoptosis of cultured human gallbladder carcinoma GBC-SD cells in vitro,so as to provide evidence for the clinical application for RES preparations and to supply new way for the treatment of gallbladder carcinoma.Methods The cultured GBC-SD cells were divided into blank control group,5-Fu positive control group,RES group,RES-BSANP group,and blank nanoparticle group.CCK-8 and RT-PCR were adopted to detect the antitumor activities in vitro,and the effects of various concentrations of drugs on the expression levels of apoptosisassociated genes of Caspase-3,Caspase-8,and Caspase-9 mRNA were investigated.Results Blank nanoparticle group showed no inhibition of GBC-SD cell growth,and 5-Fu,RES,and RES-BSANP at the concentrations of10-80μg/mL exerted the inhibition of GBC-SD cell growth in concentration-dependent manner.Compared with RES,the inhibition of the proliferative rate for GBC-SD cells by 5-Fu and RES-BSANP at the concentrations of20-80μg/mL was significantly enhanced.5-Fu,RES and RES-BSANP un-regulated the relative expression of Caspase-3,Caspase-8,Caspase-9 m RNA in GBC-SD cells,and promoted the cell apoptosis.Conclusion RES-BSANP exert a significant inhibitory effect on the proliferation of gallbladder carcinoma GBC-SD cells,and the inhibitory effect is stronger than that of RES alone.The mechanism may be related to the up-regulation of expression levels of apoptosis-associated genes of Caspase-3,Caspase-8.
