亚麻籽提取物对D-半乳糖致衰老小鼠的抗氧化保护机制研究 被引量:8

摘要 探讨亚麻籽提取物对D-半乳糖致亚急性衰老小鼠的保护作用。采用小鼠颈部皮下注射D-半乳糖的方法建立衰老模型;模型小鼠随机分为模型组及亚麻籽提取物高(600 mg/kg)、中(300 mg/kg)、低(150 mg/kg)剂量组,连续给药6周后,测定小鼠的胸腺和脾脏指数,检测小鼠全血和脑、肝、肾组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)指标。结果显示,与正常组相比,模型组小鼠脾脏指数提高、胸腺指数降低,小鼠全血和组织中MDA含量增加,SOD、CAT和GSH-Px活力降低,提示衰老小鼠模型造模成功。与模型组相比,亚麻籽提取物高剂量组能显著降低小鼠的脾脏指数,显著提高小鼠的胸腺指数,可显著降低小鼠全血和组织中的MDA含量,并能显著提高其中的SOD、CAT和GSH-Px的活力。结果表明,亚麻籽提取物对D-半乳糖所致衰老小鼠具有保护作用,其可能是通过抑制脂质过氧化,增强抗氧化酶活性发挥作用的。 The protective effect of linseed extract on D-galactose induced subacute aging mouse model was discussed.Mice were subcutaneously injected into neck to establish aging model.Successful model mice were randomly divided into model group,high(600 mg/kg),medium(300 mg/kg)and low(150 mg/kg)dose of linseed extract groups.All mice were intragastric administration for six weeks,the thymus index and spleen index were calculated,and the MDA,SOD,CAT and GSH-Px in blood,brain,liver and kidney were analyzed.The results showed that compared with normal group,the spleen index and MDA content increased,and the thymus index,SOD,CAT and GSH-Px activities decreased,which indicated that the aging mice model was successfully established.Compared with model group,the spleen index and MDA content in blood and tissue of high dose of linseed extract group decreased significantly,and the thymus index,SOD,CAT and GSH-Px activities increased significantly.These data indicated that linseed extract had protective effect on D-galactose-induced aging mouse and its mechanism may be related with the inhibition of lipid peroxides and improvment of antioxidant enzyme activities.
