摘要
目的 研究大孔吸附树脂分离富集杜仲叶中绿原酸的性能。方法 采用薄层层析 -紫外分光光度法测定绿原酸的含量 ;考察X - 5、AB - 8、S - 8和NKA - 4 4种大孔吸附树脂对绿原酸的吸附与解吸性能 ,最后选定NKA - 9树脂从杜仲叶中分离富集绿原酸 ;进一步考察进样液浓度、pH值与流速对该树脂吸附性能的影响 ,得出最佳分离条件。结果 NKA - 9树脂对提取液中绿原酸的最佳分离条件为 :当进样液浓度低于 0 .3mg·ml-1、pH 3、流速 2ml·min-1时 ,用 5 0 %乙醇洗脱 ,得到粗产品纯度为 2 5 .12 % ,收率为 78.5 %。结论 NKA - 9大孔吸附树脂可用于杜仲叶中绿原酸的分离富集。
OBJECTIVE To study the separation effect of macroporous resin for the purification of chlorogenic acid in the leaves of Eucommia ulmoides Oliv.METHODS Thin layer chromatography-UV spectrophotometry was used to analyze the content of chlorogenic acid.And the performance of adsorption and desorption for chlorogenic acid with four macroporous resins X-5、AB-8、S-8 and NKA-9 was compared.The results showed that the NKA-9 was better than the others,so the effects of concentration,pH,flow rate of the adsorbate on adsorption ability of NKA-9 resin were further researched.RESULTS The optimal extraction coditions were as follows:the concentration of extract was 0.3 mg·ml -1,pH 3 with a flow rate of 2 ml·min -1,and 50 % alcohol was used as eluant.The purity of resulted product was arrived to 25.12% and the yield was 78.5%.CONCLUSION NKA-9 resin can be used to refine the chlorogenic acid in the extraction of the leaves of Eucommia ulmoides Oliv..
