天麻对铝致大鼠学习记忆障碍的影响 被引量:14

摘要 目的 探索天麻改善铝致大鼠学习记忆障碍的作用机理。方法 将SD大鼠 36只在动物室适应性喂养一周后 ,分成 6组 ,腹腔注射生理盐水或AlCl3 溶液 0 2ml d ,每连续注射3天间歇 1天 ,6 0天内共计注射 4 5次。在间歇天肌肉注射青链霉素以抗感染。所有动物饲以普通饲料 ,自由饮水和进食 ,加天麻组 ,以 0 4g kgBW的剂量在饮水中加入天麻。每周定时称体重一次 ,调整铝 (Al)的浓度和饮水中天麻的浓度。染毒期结束后 ,用跳台实验法测大鼠的学习记忆成绩 ;然后取脑皮质 ,测定脑铝 ,做匀浆测定皮质中AChE和MAO B的活力。结果 与生理盐水组相比 ,Al3 + 5mg kg组脑铝含量升高 (P <0 0 1)、EN1增加 (P <0 0 5 )、潜伏期缩短 (P <0 0 1) ;AChE活力降低 (P <0 0 5 )、MAO活力升高 (P <0 0 1) ;Al3 + 10mg kg组脑铝含量升高 (P <0 0 1) ;EN1和EN2均明显增加 (P <0 0 1)、潜伏期明显缩短 (P <0 0 1) ;AChE活力降低 (P <0 0 1) ,MAO活力升高 (P <0 0 1)。与Al3 + 5mg kg组相比 ,Al3 + 5mg kg +天麻组大鼠EN1减少 (P <0 0 5 ) ,潜伏期延长 (P <0 0 5 ) ;AChE活力显著升高 (P <0 0 5 )、MAO活力显著降低 (P <0 0 1)。与Al3 + 10mg kg组相比 ,Al3 + 10mg kg +天麻组大鼠EN1和EN2均明显减少 (P <0 0 1) ,潜? Objective In order to investigate the mechanism of the improvement of Gastrodia elata(G.E.) on learning and memory impairment of rats exposed to aluminum.Methods 36 SD rats, adult and healthy, were divided into 6 groups at random: ①normal saline, ②Al 3+5mg/kg, ③Al 3+10mg/kg, ④normal saline + G.E., ⑤Al 3+5mg/kg + G.E. and ⑥Al 3+10mg/kg +G.E. Then, they were exposed to AlCl 3 by intraperitioneal injection at a dose of 0.2ml/d, 60d. G.E. was administered by drinking water at a dose of 0.4g/kg body weight.Results The results of learning and memory were recorded by Step-Down Test. It was showed that compared with the normal saline group, in the group of Al 3+5mg/kg, the content of brain cortex aluminum and EN1 increased significantly; the latency(LT) was shortened obviously(P<0.01); the activity of AChE decreased and the activity of MAO increased(P<0.01). In the group of Al 3+10mg/kg, the content of brain cortex aluminum, EN1 and EN2 increased significantly(P<0.01), the LT is shortened obviously(P<0.01),the activity of AChE decreased and the activity of MAO increased (P<0.01). Compared with the group of Al 3+5mg/kg, in the group of Al 3+5mg/kg+ G.E., EN1 decreased significantly(P<0.05); the LT is prolonged obviously(P<0.01),the activity of AChE increased(P<0.05) and the activity of MAO decreased (P<0.01). Compared with the group of Al 3+10mg/kg, in the group of Al 3+10mg/kg+ G.E., both EN1 and EN2 decreased significantly(P<0.01); the LT is prolonged obviously(P<0.01),the activity of AChE increased(P<0.01) and the activity of MAO decreased (P<0.01). Conclusion It is suggested that G.E. might decrease the content of brain cortex aluminum of rats. However it might improve the learning and memory ability possibly through regulating the cholinergic system and the monoaminergic system.
