艾灸“足三里”对疲劳模型大鼠海马炎症因子及小胶质细胞活化的影响 被引量:8

Effects of Moxibustion at “Zusanli(ST 36)” on Inflammatory Factors and Microglia Activation in Hippocampus of Fatigue Model Rats

摘要 目的探讨艾灸足三里治疗疲劳的可能作用机制。方法 24只SD大鼠随机分为正常组、模型组和艾灸组,每组8只。模型组和艾灸组大鼠以慢性负重力竭游泳21天建立疲劳大鼠模型。艾灸组在造模同时艾灸大鼠双侧'足三里',隔日1次,共11次;正常组和模型组不予处理。干预前后比较各组大鼠力竭游泳时间。干预后观察各组大鼠旷场实验行为,采用实时荧光定量PCR法检测海马白细胞介素1β(IL-1β)、白细胞介素6 (IL-6)、肿瘤坏死因子α(TNF-α) mRNA表达,采用免疫组化法检测海马(CA1区、CA3区)小胶质细胞标记物离子钙接头蛋白1 (Iba-1)表达。结果干预后艾灸组力竭游泳时间为(658. 00±250. 76) s,较模型组(263. 38±105. 87) s显著延长(P <0. 01)。各组大鼠旷场实验的水平移动距离、垂直移动距离差异均无统计学意义(P> 0. 05)。与正常组比较,模型组大鼠海马IL-1β、IL-6、TNF-αmRNA表达及Iba-1表达升高(P <0. 01);与模型组比较,艾灸组大鼠海马IL-6 mRNA表达及Iba-1表达降低(P <0. 05或P <0. 01)。结论艾灸'足三里'可改善大鼠运动性疲劳,其机制可能与抑制海马小胶质细胞的活化和炎症因子有关。 Objective To explore the possible action of moxibustion at Zusanli(ST 36)in treating fatigue.Methods A total of 24 SD rats were randomly divided into a normal group,a model group,and a moxibustion group,with 8 rats in each group.Rats in the model group and the moxibustion group were exposed to chronic loaded gravity for 21 days to establish a fatigue rat model.In the moxibustion group,moxibustion on bilateral'Zusanli(ST36)'was performed,once every other day for 11 times.The normal group and the model group were not treated.The exhaustive swimming time of each group was compared before and after the intervention.After the intervention,the open-field experimental behavior of the rats in each group was observed.Real-time quantitative polymerase chain reaction(PCR)was used to detect the mRNA expression of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in hippocampus.Immunohistochemistry was used to detect the expression of microglia marker ion-calcium adaptor protein 1(Iba-1)in hippocampus(CA1 region,CA3 region).Results After intervention,the exhaustive swimming time of the moxibustion group was(658.00±250.76)s,which was significantly prolonged than that of the model group(263.38±105.87)s(P<0.01).There was no significant difference in the horizontal movement distance and vertical movement distance of the open-field test between the groups(P>0.05).Compared with the normal group,the expression of IL-1β,IL-6,TNF-αmRNA and Iba-1 in the hippocampus of the model group was increased(P<0.01).Compared with the model group,the hippocampus IL-6 mRNA expression of the moxibustion group and Iba-1 expression were decreased(P<0.05 or P<0.01).Conclusion Moxibustion at'Zusanli(ST 36)'could improve exercise-induced fatigue in rats,and its mechanism may be related to inhibition of hippocampal microglial activation and inflammatory factors.
