摘要
目的观察菖欢胶囊对慢性利血平孤养抑郁性神经症模型大鼠行为的影响。方法选择Open-Field法评分相近的50只成年SD雄性大鼠,随机分为对照组、模型组、菖欢胶囊小、大剂量治疗组和阿米替林治疗组,每组10只。应用分养和长期腹腔注射利血平造成大鼠抑郁性神经症模型,以Open-Field法和糖水消耗实验对照研究抑郁性神经症大鼠行为的改变。结果与对照组比较,抑郁模型组大鼠体重下降,Open-Field法评分降低,糖水消耗量减少;与模型组比较,菖欢胶囊大剂量组和阿米替林组大鼠体重增加,菖欢胶囊小剂量组体重无明显变化,菖欢胶囊大剂量组Open-Field法水平运动和垂直运动评分均升高,菖欢胶囊小剂量组和阿米替林组对大鼠水平活动无明显影响,垂直运动评分升高,各组大鼠糖水消耗量均增加。结论菖欢胶囊可明显改变慢性利血平和孤养诱导的抑郁性神经症模型动物的抑郁行为,其抗抑郁作用机理可能与改善中枢单胺能神经的功能状态有关。
Objective To investigate the effect of ChangHuan capsule on behavior in the separation and chronic reserpine-induced depressive neurosis rat model. Methods Male Sprague-Dauley rats were randomly allocated to five groups as follows:control, model, amitriptyline, ChangHuan capsule Ⅰ (general dose) and ChangHuan capsuleⅡ(overdose). The depressive neurosis model of rat was produced by separation and chronic reserpine injection.The behavior of rat was detected by Open-Field test. Open-Field and sucrose consumption were used to study the chandges of rat behavior. Result It showed that the weight, Open-Field marks and sucrose consumption are lower in the model, and the weight are higher in the ChangHuan capsule Ⅱ and amitriptyline. Crossing marks is higher in the ChangHuan capsuleⅡ. Rearing marks and sucrose consumption are higher in the amitriptyline, ChangHuan cappsuleⅠand ChangHuan capsuleⅡ. Conclusion These data suggest that ChangHuan capsule was effective in the treatment of depressive neurosis. It antidepressive mechanism might be related to balance the function of Central Monoaminergic nervous system.
