摘要
没食子蓝在239 nm紫外光激发下,在414 nm处出现一个小的荧光峰,加入小牛胸腺脱氧核糖核酸(ctDNA),该荧光峰的强度大大增强。其荧光增强比值与ctDNA的浓度分别在低浓度区间(0.05~1 mg/L)和稍高浓度区间(1~10 mg/L)是较好的线性关系,利用这一原理建立了一种用没食于蓝作为荧光探针测定DNA的方法。该方法相当简单,其检测限可达0.04mg/L。
The fluorescent enhancement of gallocyanine by nucleic acid in aqueous solution was studies. Gallocyanine and nucleic acid interacted strongly with each other. The excitation wavelength was 239nm and the emission wavelength was 414 nm. The intercalation mole was prove to exist. The calibration curve was linear in the range of 0.05 - 1mg/L and 1-10 mg/L for calf thymus DNA. The limit of determination was 0.04mg/L.
