摘要
目的 :建立灵敏、准确、简便、快速的方法测定人尿中氟罗沙星的含量。方法 :以 2 90nm为激发波长 ,以 458nm为发射波长 ,用荧光分光光度法测定人尿中氟罗沙星的排出速率。结果 :氟罗沙星浓度在 0 .0 5~ 0 .5mg·L- 1 范围内 ,荧光强度与浓度呈良好的线性关系 ,检测限为 0 .0 3mg·L- 1 。精密度为日内RSD <1 .1 0 % ,日间RSD <2 .98% (n =5) ,回收率为 (98.6±1 .3) %。结论 :采用此法测定了 2名健康受试者口服氟罗沙星片后的尿药排出速率。尿中药物排出速率最高是在服药后 6~ 8h ,2 4h内尿药累积排出率为 56 .1 2 % ,0~ 2 4h内尿药浓度为 (69.3± 40 .6)mg·L- 1 ,显著高于大多数常见致病菌的MIC90 ,表明有效尿药浓度至少可维持 2 4h。
OBJECTIVE To develop a new method for determination of fleroxacin in human urine. METHODS A fluorescence spectrophotometry was applied with excitation wavelength of 290 nm and emission wavelength of 458 nm.RESULTS The standard curve was linear in the concentration range of 0.05 to 0.5 mg ·L -1 ,the detection limit was 0.03 mg ·L -1 , the average recovery was 98.6% . CONCLUSIONS The excretion rate of fleroxacin in urine for two healthy volunteers after a single oral dose of 200mg fleroxacin tablets was determined by this method. The result can provide basis for clinical application of fleroxacin.
