摘要
目的 :建立牛血清白蛋白手性柱直接拆分亚叶酸钙消旋体的方法。方法 :采用高效液相色谱法 ,使用 EC15 0 / 4RESOLVOSIL BSA- 7(15 0 mm× 4 mm)手性色谱柱 ,0 .2 0 mol/ L、p H5 .0磷酸盐缓冲液为流动相 ,对亚叶酸钙消旋体进行拆分 ,考察了流动相缓冲液浓度、p H值以及柱温对拆分效果的影响。结果 :流动相的缓冲液浓度、p H和柱温对亚叶酸钙对映体的容量因子 k′,以及分离度 Rs,均有较大影响。左旋亚叶酸钙和右旋亚叶酸钙分别在18.5 min和 2 2 .6 min出峰 ,分离度 Rs=1.4 9。结论 :用本方法可以成功拆分亚叶酸钙对映体。
Objective: To establish a direct and fast method separating calcium levofolinate and calcium dextrofolinate in a bovine serum albumin stationary phase chiral column. Methods: Using EC 150/4 RESOLVOSIL BSA-7 (150 mm×4 mm) chiral separation column,with 0.20 mol/L, pH=5.0 phosphate buffer as mobile phase,HPLC method was performed to separate calcium folinate enantiomers. Results: The capacity factor and resolution of the two calcium folinate enantiomers were greatly affected by mobile phase buffer concentration,pH and the column temperature. And the retention time of calcium levofolinate and calcium dextrofolinate were 18.5 min and 22.6 min, respectively. The resolution, Rs=1.49. Conclusion: Calcium folinate enantiomers are separated successfully using this method.
