摘要
本文报告了茯苓素对人急性早幼粒白血病细胞系HL-60增殖的抑制作用.经25～100μg/ml茯苓素处理1天,HL-60细胞DNA合成能力明显减弱.药物作用2天后,细胞计数明显少于对照组.这种效应随药物浓度的加大及作用时间的延长而增强.半数抑制浓度为58.4μg/ml.抑制作用为不可逆性.茯苓素处理4天后,HL-60细胞自发形成集落的能力明显减弱.细胞周期分析结果,加药后G_0/G_1期细胞明显增多,S和G_2+M期细胞减少.
The present study demonstrated the inhibitory effect of poriatin on proliferation of human promyelocytic leukemic cell line HL-60.Treated with poriatin at concentration of 25-100μg/ml,The incorporation of ~3H-TdR into HL-60 cells reduced 14.1-77.3% while the cell number decreased afterward in a dose and time dependent manner.The inhibitory effect was irreversible.IC_(50)=58.4μg/ml.The suppression of DNA synthesis became evident on day 4.Moreover,the ability of spontaneous colony formation was significantly decreased.The histogram of DNA distribution analyzed by flow cytometry also demonstrated a higher percentage of HL-60 cells in G_0/G_1 phase after poriatin treatment while the cells in S and G_2+M phases decreased remarkably.
