DETERMINATION OF PLATINUM IN HUMAN PLASMA URINE AND HYDROTHORAX BY GRAPHITE FURNACE ATOMIC ABSORPTION SPECTROMETRY

摘要 本文研究了石墨炉原子吸收光谱法测定顺氯氨铂在人的血浆、胸水及尿中铂浓度的方法.样品无需预先处理,加入Triton X-100后,可直接进样测定.测定可以对照酸标准液,不需加入空白血浆,其方法简便、灵敏度高、重复性好,适用于临床生物样品的测试.检出限5ppb,绝对灵敏度为7×10^(-11)g/0.0044A,线性范围50~800ng/ml,变异系数四个浓度为1.93~2.18%(n=10),回收率5个浓度为96.1~107.6%. A method is described for the determination of platinum in human plasma,urine and hydrothorax following therapeutic dosing with cisplatin.The main advantages of this method are simple,sensitive and precise.Analyses are performed after adding triton X-100,without sample preparation.The measurements can be made against aqueous standards.This method is therefore suitable for routine drug monitoring.The limit of detection was about 5 ppb.The absolute sensitivity was 7×10^(-11)g/0.0044 A.The linear range was 50-800 ng/ml.The precision of the assay (CV) was 1.93-2.18%(n=10).The recovery was 96.1-107.6%.
