地佐环平对缺氧缺糖海马脑片CA1区Bcl-2蛋白和Bax蛋白表达的影响 被引量:2

摘要 目的:观察N-甲基-D-门冬氨酸(NMDA)受体非竞争性拮抗剂MK-801对缺氧缺糖海马脑片CA1区Bcl-2蛋白和Bax蛋白表达的影响。方法:新生6~9dSD大鼠断头取脑,制备海马脑片,将各孔脑片单纯随机分为3组,分别为正常对照组(HOTC)、单纯缺氧缺糖组(OGD)和预加MK-801再缺氧缺糖组(MK-801+OGD)。海马脑片进行缺氧缺糖培养,最后进行免疫组织化学染色和图像分析处理。结果:各组海马脑片CA1区均有不同程度Bcl-2,Bax蛋白表达。OGD组Bcl-2蛋白的表达(15.7±1.1)弱于HOTC组(18.5±0.9)和MK-801+OGD组(18.0±0.9),差异均有显著性意义(F=6.495,P<0.05);OGD组Bax蛋白的表达(13.1±2.5)强于HOTC组(7.3±1.1)和MK-801组(8.7±1.1),其差异均有显著性意义(F=9.770,P<0.05)。同时,OGD组CA1区出现细胞缺失形成的空洞,其他两组与OGD组相比有所减少。结论:MK-801可增强缺氧缺糖海马脑片CA1区Bcl-2蛋白的表达,降低Bax蛋白的表达,并减轻该区锥体细胞损伤程度。 AIM:To investigate the effect of N methyl D aspartate(NMDA) recipient non competitive antagonist MK 801 on the expression of Bcl 2 and Bax proteins in the CA1 subfield of the hippocampal organotypeic tissue cultures(HOTC) undergoing oxygen/glucose deprivation(OGD). METHODS:SD rats,6-9 days old,were sacreficed for brain,HOTC were made and randomly divided into 3 groups:normal control(HOTC) group,OGD group and MK 801+OGD group.HOTC were undergone OGD,immunohistological staining and image analysis were made at last. RESULTS:There were different expression of Bcl 2,Bax proteins in the CA1 subfield of the three groups.The expression of Bcl 2 protein in the OGD group(15.7±1.1)was weaker than that in the HOTC group(18.5±0.9)and the MK 801+OGD group(18.0±0.9),the differences were significant(P< 0.05,F=6.495).The expression of Bax protein in the OGD group(13.1±2.5)was stronger than that in the HOTC group(7.3±1.1)and the MK 801 group(8.7±1.1),and the differences were significant(P< 0.05,F=9.770)And there were the vacum caused by cell depletion in the CA1 subflied in the OGD group,the vacum in the CA1 subflied in the MK 801 group was fewer than that in the OGD group. CONCLUSION:MK 801 can enhance the expression of Bcl 2 protein and weaken the expression of Bax protein in the CA1 subfield of oxygen and glucose deprived hippocompical organotypic tissue culture of rat,and reduce the injury degree of pyramidal cell in the subfied.
