摘要
该文根据糖化酶能水解淀粉的性质,采用Search-PAGE在分离胶中加入淀粉作为糖化酶的作用底物,电泳结束后温浴一定时间,糖化酶所在部位胶板中的淀粉被酶解,其余部分的淀粉依然存在,碘液染色后,糖化酶存在的部位呈现透明区域,没有被水解的部位则被染成蓝色,每块胶板1次可筛选出8~15个样品.
PAGE was run by adding a certain concentration of starch into polyacrylamide gel. The band of saccharifying enzyme could be detected easily because of its hydrolysis of starch, and the active band showed transparent and the other area was all blue after staining by iodine. A simple, sensitive and convenient method of detecting saccharifying enzyme was established, and a great deal of 8-15 samples could be screened with one gel in a shorter time.
