植物NAD-IDH基因克隆及体外表达酶活力分析

摘要 植物NAD+-依赖型异柠檬酸脱氢酶(NAD-IDH)是一多功能酶.以拟南芥生态型“Columbia gll”及甘蓝型油菜三系“陕2A”,“陕2B”和“垦C1”的叶片为材料,采用RT-PCR方法成功地从每种植物材料中分别克隆了3种NAD-IDH不同亚基基因.同源性搜索发现,来源于油菜的克隆基因是新基因.将克隆基因的功能蛋白编码区整合到pMID1多克隆位点,构建表达重组体,均能在体外翻译系统中高效地表达出特异目的蛋白.亚基0,亚基1和亚基2基因转录本加入到含终浓度为36μg/mL的二硫键异构酶的体外翻译系统,体外表达产物中检测到NAD-IDH酶活力.不同基因种类及转录本分子比的体外表达产物的酶比活力比较表明,NAD-IDH由3种亚基组成,缺一不可,缺少亚基0是致命的,缺少亚基1或2中的任何一个对酶活力的损伤是严重的.同时发现,来源于陕2A和陕2B的亚基1基因转录本中存在碱基缺失现象,从而发生移框突变或无义突变,使突变亚基不表现正常亚基1功能,导致油菜品系间叶片NAD-IDH酶比活力存在差异.
