目的 观察转入野生型 PTEN基因的胶质瘤细胞体外侵袭力改变 ,探索 PTEN基因对胶质瘤细胞影响方式。方法 构建野生型 PTEN基因的 pc DNA3 .1Hygro(-)真核质粒载体 ,并用于转染 PTEN基因失活的U 2 51细胞系 ,采用重组基底膜侵袭模型 ,观察转染前后细胞侵袭力的改变。结果 转染 PTEN基因可以明显抑制U2 51细胞的侵袭力。结论

ObjectiveTo study the effects of wild-type PTEN gene on the invasion ability of glioma c ells. MethodsA pcDNA3 1/Hygro(-) recombinant plasmid containi ng PTEN gene was constructed. U 251 cells which express no PTEN were transf ected with wild-type PTEN plasmid vector and stable transfected clones were sel ected. The invasion ability of transfected cells was observed. Results The invasion ability of the transfected cells was markedly inhibited. ConclusionWild-type PTEN gene could inhibit the invasion abil ity of glioma cells.
