摘要
目的 :选择空白囊泡制备的最优条件。方法 :采用冷冻干燥法制备空白囊泡 ,以正交设计法对冷冻时间、冷冻方式、水和叔丁醇的用量等影响囊泡质量的 4种冻干因素进行优选。结果 :分析得出在冷冻室温度和真空度恒定条件下 4种考察因素的最优条件 :冷冻时间对囊泡成型和稳定性的影响最小 ,可作为误差项 ,实际操作中时间应大于 3 0分钟。冷冻方式以搅拌或速冻方式较好 ,根据实际情况以速冻方式较为简便。水和叔丁醇用量影响程度较大 ,水量最佳条件为最大值 (0 2mL) ,叔丁醇量最佳条件为最小值 (5mL) ,按优化条件重复试验 ,得到的结果与预测值一致。结论 :优选出的空白囊泡冻干制备条件 ,具有操作简便 。
Objective:To study the optim um conditions of empty niosomes prepared by lyophilization. Methods: The empty niosomes were prepared by lyophilization. The optimum freeze-dry conditions were selected by orthogonal design. Results:The opti mum freeze-dry conditions of the empty niosomes were:at least 30 min of freezin g time, rapid freezing, distilled water of 0 2 mL and tert-butanol of 5 mL. A sample with the best value has been produced with lyophilization. Conc lusion:The optimum conditions of empty niosomes prepared by lyophi lization are practicable and successful.
