摘要
目的 :探讨脑复苏后神经细胞凋亡的相关基因的调控机制。方法 :大鼠脑缺血再灌注损伤后 ,分别应用TUNEL染色法和免疫组化法检测海马CA1 区凋亡的神经细胞和Bcl- 2、Bax、C -fos、Fas、p53、Capases 3蛋白在细胞中的表达。 结果 :与假手术组相比 ,缺血再灌注 2 4h后 ,凋亡的神经细胞增加 (P <0 0 1 ) ,Bcl- 2蛋白表达的阳性细胞减少 (P <0 0 1 ) ,Bax、C -fos、Fas、p53、Capases 3蛋白表达的阳性细胞增加 (P <0 0 1 )。结论 :脑复苏后 ,多基因参与了神经细胞凋亡的调控 ,Bcl- 2具有抗凋亡作用 ,Bax、C -fos、Fas、p53、Capases 3蛋白表达增加可促进细胞凋亡。
Objective:To investigate the mechanisms of gene and neuron apoptosis after cerebral resuscitation.Methods:TUNEL stain and immunohistochemistry stain were applied for 102 mature Wistar rats of ischemia-reperfusion to determine the neuron apoptosis positive cells and the protein expression positive cells of Bcl-2,Bax,C-fos,Fas,Capases 3,p53 respectively.Results:Compared with the sham-operated control group,the ratio of the neuron apoptosis positive cells and the protein expression positive cells of Bax,C-fos,Fas,Capases 3,p53 increased significantly(P<0.01),the ratio of the protein expression poression positive cells of Bcl-2 decreased significantly(P<0.01).Conclusions:After cerebral resucitation,multigene regulate neuron apoptosis,Bcl-2 expression might promote the neural survival,and that of Bax,C-fos,Capases 3,Fas,p53 would enhance the neuron apoptosis.
