摘要
目的 :观察水蛭注射液对大鼠脑缺血再灌注损伤的防治作用,并初步探讨其作用机制。方法 :采用大鼠双侧颈总动脉结扎再灌注模型 ,检测大鼠脑组织含水量及Ca2 + 含量的变化 ,脑组织匀浆及血清中的MDA ,SOD ,NO的含量 ;病理切片HE染色观察组织病理学变化。结果 :水蛭注射液能显著降低脑缺血再灌注损伤大鼠脑组织含水量及Ca2 + 含量 ,降低血清及脑组织中MDA ,NO的含量 ,提高SOD的活性。细胞形态学变化显示该药物对神经细胞的保护效果优于对神经胶质细胞的保护效果。结论 :水蛭注射液对大鼠脑缺血再灌注损伤有一定的预防和治疗作用 ,与药物剂量大小关系不密切 ;对大鼠大脑灰质的防治效果比对大鼠大脑白质的防治效果好。
Purpose:To observe the protective and curative effects of Shizhi(leech)Injection on cerebral ischemia/re-perfusion injury in rats and to explore its mechanism.Methods:The model rat was prepared by ligature to determine the water content and Ca 2+ content in the rat brain tissue and also MDA,SOD,NO contents in brain homogenate and blood serum.The changes of histopathology were observed by the HE staining method.Results:Compared with the control group,the injection could greatly lower the water contents,Ca 2+ contents in the rat brain tissue and MDA and NO contents in brain homogenate and blood serum and increase SOD activity in the treatment group.The changes in cytomorphology indicated the better protective action on nerve cells than on neuroglia cells.Conclusions:Shuizhi Injection can play a good role in preˉventing and treating the celebral ischemia/reperfusion injury in rat without close relevance to the drug dosage.And better effects can be achieved in prevention and treatment of brain cinerea than alba.
