新疆紫草外植体诱导愈伤组织研究

STUDY ON THE CALLUS WERE INDUCED FROM SEGMENTS OF Arnebia euchroma(Royle)Johnston

以新疆紫草(Arnebia euchroma(Royle)Johnston)幼叶、子叶和胚轴作为外植体,诱导产生了愈伤组织,幼叶和子叶在MSB+NAA0.4ppm+2.4—D0.2ppm+6—BA0.6ppm的琼脂培养基上脱分化效果好,愈伤组织为白色半透明状;胚轴在LS+NAA0.4ppm+IAA1.5ppm+KT2.0ppm的琼脂培养基上脱分化较合适,愈伤组织为红、白两种颜色,平均脱分化率为68.7%,这些愈伤组织有紧密型和松散型两种形态,扫描电镜观察了紧密型白色愈伤组织的表面结构,可看到愈伤组织表面有许多突起的胚性细胞团,在继代培养过程中,多数这种愈伤组织能够再分化,形成胚芽。

Callus were inducded from sogments of leaflet, cotyledon and plumular axis of the Arnebiaeuchroma(Royle)John(?)on it(?) right for the leafler and cotyledon to be culturde on MSB agarmedium, supplemented with NAA0.4ppm+2.4--D0.2ppm+6--BA0. 6ppm. The calli arewhite translucent. It is suitable for plumular axis on the LS agar medium,with NAA0.4ppm+IAA1. 5ppm+KT2.0ppm. The calli are the red and white. The average frequency of dedif-fer(?)ntiation was 68.7%. There callus have two types, tight and loose. The surface structureof the tight white callu was observed by scanning electron microscope, there are many embry-onic cells projecting in surface of the callu can be seen. The majority of these calli can formembayonic bud after redifferentiation in the course of continuous culture.
