人neurotrimin基因的克隆及初步功能研究 被引量:1

摘要 在神经系统的发育过程中,免疫球蛋白超家族成员参与多种特异的细胞与细胞间相互作用。为了发现及研究这个家族在人神经系统中的新成员,对人脑cDNA文库进行筛选,得到一全长cDNA克隆。该cDNA含一个1032bp的开放阅读框,编码一个含344个氨基酸的蛋白质。序列分析发现它是一个糖蛋白,含3个C_2样免疫球蛋白结构域,通过GPI锚定到膜上。同源性比较发现,该蛋白质与大鼠neurotrimin(Ntm)有97%的同源性,因此将它命名为人neurotrimin(NTM)。Northern杂交发现它有3.2,4.0和9.0kb3个转录本。值得注意的是,NTM在胎脑中的表达高于成年脑,在神经肿瘤中的表达高于正常脑组织。将HA表位插入第3个免疫球蛋白结构域与GPI信号之间,构建入真核表达载体pcDNA3.1+/Zeocin中,转染CHO细胞筛选出稳定表达的细胞株。凝集实验发现,HA-NTM表达于CHO细胞的表面并能加强细胞间的凝集。
