抑制消减杂交结合cDNA微阵列技术鉴定人肾癌组织高表达基因谱 被引量:8

摘要 为克隆肾癌组织高表达基因,探讨肾癌发生、发展的分子基础,以人肾癌组织mRNA为试验方,正常肾组织mRNA为对照构建抑制消减杂交文库。用文库379个克隆制作基因芯片,以同位素标记探针对文库进行高通量筛选,对肾癌组织中表达增高6倍以上的67个克隆进行测序,结果在GenBank中进行比对分析,其中4个克隆为新EST片段,2个为KIAA类基因,其余克隆为已知基因。其中已有文献报道的肾癌相关基因,如VEGF,波形蛋白,组织因子等,大部分为新发现的肾癌相关基因,包括锌带蛋白、垂体瘤转化基因1等。3个EST及一些在肾癌组织中高表达的已知基因以Northern杂交、RT-PCR作了验证。免疫组化与Western blot进一步证实锌带蛋白与垂体瘤转化基因1在肾癌组织中表达显著增强。这些高表达的基因可能成为肾癌诊断、治疗的潜在靶点,有助于从基因谱的角度进一步认识肾癌发生、发展的分子机制。
