摘要
以40%饱和度的硫酸铵溶液去除杂蛋白,用80%饱和度的硫酸铵和75%的丙酮纯化米曲霉β—半乳糖苷醇,其比活力为147.7U/mg,回收率为38.1%。该酶最适反应温度和pH分别为50~55℃和4.0~6.0,作用于乳糖的米氏常数K_m为16.7mmol/L,最大反应速度V_(max)为5.56μmol/min。
The β-galactosidase from aspergillus oryzae was purified by centrifuging, ammonium sulfate precipitation and acetone treating. The activity of purified β-galactosidase was 147. 7μ/mg. The recovery was 38. 1%. The optimum reactive temperature and pH for the enzyme were 50~55℃ and pH4. 0~ 6. 0,the Km for lactose was 16. 7μmol/L and Vmax was 5. 56umol/min
