摘要
研究了以中药渣为原料培养灵芝固体菌质,从中提取灵芝多糖的工艺条件和影响苯酚—硫酸法测定灵芝多糖的因素,结果表明:490nm,25℃,25min,1.6mL 6%苯酚,7.5mL浓硫酸为苯酚—硫酸法的较优测定条件,料水比1:60,85℃,2~2.5h为提取灵芝多糖的最佳工艺条件,此条件下测定甜芝、韩芝和赤芝固体菌质中灵芝多糖含量分别为4.24%,3.35%,3.11%,子实体中多糖含量分别为1.49%,1.34%,1.58%,经TCL鉴定,提取的菌质多糖确为灵芝多糖。
A study is made on producing Ganoderma lucidum fungal substance with herb residues as raw material and extracting Ganoderma lucidum polysaccharide (GLP) from them, and phenol-sulfuric acid method was used to determine the content of GLP. Through experiments we optimized the measuring conditions of phenol-sulfuric acid method (Amax= 490nm,25C ,20min,l. 6mL 6% phenol,7.5mL sulfuric acid) and extracting conditions of GLP (60 times water ,85C ,2~2. 5h). The content of GLP is respectively 5. 24%(Sweet Ganoderma lucidum) ,3. 35%(Korea Ganoderma lucidum) ,3. 11%(Red Ganoderma lucidum) in medicinal fungal substance and 1. 49%, 1. 34%, 1. 58% in corresponding fruit bodies. Through TLC identification, we made ascertain the polysaccharide extracted from fungal substance was GLP.
