摘要 目的 :研究白介素 10 (IL 10 )对人滋养层细胞中人类白细胞抗原 G(HLA G)mRNA表达及人绒毛膜促性腺激素 (HCG)分泌的影响 ,探讨IL 10在母 -胎免疫耐受机制中的作用。方法 :选取正常妊娠 6~ 8周妇女行人工流产术后的绒毛组织 2 1例 ,每 1例标本随机平均分为实验组和对照组 ,加入IL 10培养和单独培养 72小时 ,每组均采用逆转录 -聚合酶链式反应 (RT PCR)及放射免疫测定 (RIA)的方法分别检测HLA GmRNA的表达量及HCG的分泌水平。结果 :①两组滋养层细胞中均有HLA GmRNA的表达及HCG的分泌 ,实验组HLA GmRNA的平均表达峰度值为 0 2 8± 0 0 7,对照组HLA GmRNA的平均表达峰度值为 0 15± 0 0 4 ,差异有非常显著性 (P <0 0 1) ;实验组HCG浓度为 2 7 2 0± 4 16U/L ,对照组HCG浓度为 2 3 98± 2 2 7U/L ,差异有非常显著性 (P <0 0 1)。②直线相关分析表明 ,滋养层细胞中HLA GmRNA的表达与HCG分泌之间无相关性。结论 :IL 10诱导人滋养层细胞中HLA GmRNA的表达增高及HCG的分泌增加 ,对促进母 Objective:To investigate the effects of IL-10 on the expression of HLA-G mRNA and the secretion of HCG in human trophoblasts, and to study the function of IL-10 in the mechanism of maternal-fetal immune tolerance.Methods:21 trophoblast explants were obtained from induced abortion of normal pregnancies at 6 to 8 weeks of gestation. Trophoblast explants originating from the same sample were incubated for 72h in two groups, one with IL-10 and the an other without IL-10. The reverse transcription-polymerase chain reaction (RT-PCR) was used to analyze the expression of HLA-G mRNA, and the radioimmunoassay (RIA) was used to analyze the secretiory level of HCG. Results:①The mRNA of HLA-G and HCG were found in every sample. The average levels of HLA-G mRNA of two groups were 0.28±0.07 and 0.15±0.04, respectively. The difference between the two groups was significant ( P <0.01). The average levels of HCG of two groups were 27.20±4.16 U/L and 23.98±2.27 U/L, respectively. The difference between the two groups was significant (P<0.01). ②There was no correlation between the expression of HLA-G mRNA and the secretion of HCG in human trophoblasts.Conclusions:IL-10 can enhance the expression of HLA-G mRNA and the secretion of HCG in trophoblasts, and play a important vole to promote the formation of maternal-fetal immune tolerance.
