摘要
目的 :研究丹参对兔重症急性胰腺炎 (SAP)早期中性粒细胞 (PMN)与内皮细胞 (EC)粘附作用及对血浆肿瘤坏死因子 α(TNF α)的影响。方法 :将日本大耳兔 2 4只建立兔SAP模型 ,随机分为丹参治疗组和对照组。分别于术前 ,术后即刻、2、4、8、16h ,取血分离获得PMN。与体外培养的兔血管EC作用测定PMN EC粘附率 ,并测定各时点血浆TNF α值。结果 :两组术后即刻粘附率均升高 ,对照组术后 2hTNF α开始升高 ,8h达高峰 ,16h仍高于正常 ;治疗组 2、4、8、16h各时点的TNF α的值小于对照组 (P <0 .0 5 )。结论 :丹参能降低SAP血浆TNF α的水平 ,进而抑制PMN EC粘附 。
Objective:To study the effect of salvia miltiorrhiza on the adhesion of polymorphonuclear leukocyte(PMN) and endothelial cells(EC) and on TNF α in rabbits with severe acute pancreatitis(SAP) of early stage. Methods: Twenty four Japan rabbits were induced SAP and divided randomly into control group and salvia miltiorrhiza treatment group. Blood was obtained respectively before operation, instantly after operation, 2 hours, 4 hours, 8 hours, 16 hours after operation. PMN was separated from the blood and interacted with rabbit's vascular EC cultured in vitro. The adhesion ratio of PMN and EC and expression of TNF α was measured. Results: After operation, the adhesion ratio of the two groups both rose. In the control group TNF α began to rise 2 hours after operation, reached the peak 8 hours after the operation, and was above normal 16 hours after operation. In the treatment group 2 hours, 4 hours, 8 hours, 16 hours after operation, TNF α was lower than the control group (P<0.05). Conclusion: Saliva miltiorrhiza can lower the TNF α level of SAP of early stage, thus inhibit the adhesion of PMN and EC, improve the microcirculation and ease the tissue damage.
