诱导人骨髓间充质干细胞向软骨细胞分化的实验研究 被引量:3

摘要 目的 探讨人骨髓间充质干细胞 (mesenchymalstemcells,MSCs)向软骨细胞诱导分化的能力。方法 选用不同代次的MSCs,使用条件培养基 ,观察细胞的形态变化 ,同时采用细胞化学和免疫组化的方法检测糖胺多糖的分泌和Ⅱ型胶原的表达。结果 MSCs在转化生长因子 -β1 、维生素C的作用下 ,7天后可见细胞多变为圆形或随圆形 ,糖胺多糖和Ⅱ型胶原染色阳性 ,表现出软骨细胞的分化特点。结论 在一定培养条件下成功诱导人MSCs向软骨细胞分化。MSCs作为骨组织工程学方面的种子细胞具有潜在的应用价值。 Objectives To investigate the chondrogenic differentiation ability of human bone marrow mesenchymal stem cells(MSCs). Methods After adherent cell cultures were established in vitro condition, we observed the morphological changes of human bone marrow MSCs under chondrogenic induction. The expression of type-Ⅱ collagen and glycosaminoglycans were detected by immunohistochemical and cytochemical assay in extracellular matrix. Results MSCs became round or ovoid from spindle shape after 7 days exposure to TGF-β1 and vitamin C in conditional culture. Histological analysis of the cells revealed the characteristics of chondrogenic differentiation with expression of type-Ⅱ collagens and abundance of glycosaminoglycans within the extracellular matrix. Conclusions MSCs derived from human bone marrow may be induced into chondrocytes under conditional culture media in vitro, which may serves as the potential source of chondrogenic seed cells for bone tissue engineering.
