摘要
目的 探讨红花在大鼠全脑缺血再灌注损伤中的脑保护作用及其分子机制。方法 选用Wistar大鼠 70只 ,随机分为红花组 ,盐水对照组 ,假手术组 ,单纯缺血组。制备大鼠全脑缺血再灌注损伤模型。通过光镜、电镜、原位杂交及免疫组化方法测定Caspase 3、Bcl 2在脑缺血再灌后不同时间蛋白表达的变化 ,并用图像分析方法测定其免疫强度。结果 红花与盐水对照组相比使相应时间点Caspase 3表达降低 ,Bcl 2表达升高。结论 红花可减轻脑缺血再灌注损伤 ,具有脑保护作用。
Objective To explore the protections and molecular mechanism of safflower in rat cerebral ischemia reperfusion. Methods 70 Wistar rats were randomly allocated to four groups:safflower group, normal saline control group, sham-operation group, ischemia-only group. To make the model of global cerebral ischemia reperfusional injury rat. By light microscope, electron microscope, immunohistochemical method and hybridization in situ technique to observe the expression of Bcl-2 and Caspase-3 after cerebral ischemia reperfusion varing with time and to measure the expression levels of Bcl-2 and Caspase-3 by graphic analysis.Results The expression of Caspase-3 is lower and Bcl-2 is higher in safflower group than those in normal saline control group at the same time. Conclusion Safflower can alleviate the injury of global cerebral ischemia reperfusion.
